摘 要:目的建立HPLC法测定瓶尔小草中槲皮素含量的方法。方法采用Kromasil 100-5-C18色谱柱(250mm×4.6mm,5μm),流动相为乙腈(A)-0.2%醋酸水(B)(27∶73)等度洗脱,检测波长为360nm,流速为1.0m L·min-1,柱温:30℃。结果槲皮素进样量在26.78~178.56ng范围内与峰面积呈良好线性关系(r=0.9997,n=6),槲皮素的平均回收率为100.31%(RSD=1.204%)。结论该法简便、准确、可靠,可用于瓶尔小草中槲皮素的含量测定。运用该法测定不同干燥方法的瓶尔小草样品中槲皮素含量,结果二者槲皮素含量差距较大,低温烘干样品中槲皮素含量明显高于阴干样品。OBJECTIVE To establish a method for the quantification of quercetin in Ophioglossum vulgatum L. by HPLC. METHODS The experimental conditions list as follow Kromasil 100-5-C18(250 mm × 4. 6 mm,5μm)column,mobile phase of acetonitrile-acetic acid water(27:73,p H = 2 ~ 3),30℃,flow rate of 1. 0 m L·min^-1,detection wavelength of 360 nm. RESULTS The method had good linear relationship within the range of 26. 78 ~178. 56 ng( r = 0. 9997) for quercetin. The average recoveries( n = 6) were 100. 31%( RSD = 1. 204%) for quercetin. The contents of quercetin in Ophioglossum vulgatum L. by dring in the shade. and dring in low temperature were0. 0194% and 0. 0404% respectively. CONCLUSION The content of quercetin in Ophioglossum vulgatum L. by different drying methods. was different. This method is convenient,accurate and reliable for the quantification of quercetin in Ophioglossum vulgatum L.
