摘 要:目的通过建立血管内皮损伤模型,研究蓝莓原花青素抗氧化应激血管损伤的机制。方法索氏提取法提取蓝莓果中原花青素,用过氧化氢添加法建立细胞氧化模型,添加不同浓度的蓝莓原花青素和原花青素标准品,采用流式细胞术分析内皮细胞活力、凋亡和线粒体功能的变化。结果 H_2O_2诱导后细胞内ROS增加并且细胞发生显著的凋亡和坏死,原花青素标准品和蓝莓原花青素提取物与H_2O_2联合处理后,细胞内ROS有降低的作用,坏死细胞的百分率有所降低。结论蓝莓原花青素具有一定抗氧化性,并且细胞毒性比较弱。Objective To establish a vascular endothelial injury model to study the anti-oxidative stress of blueberry procyanidins vascular damage mechanism. Methods Soxhlet extraction of procyanidins from blueberry, cell oxidative model was established by adding hydrogen peroxide, add different concentrations of blueberry proanthocyanidins and proanthocyanidins standards, flow cy- tometry was used to analyze the changes of endothelial cell viability, apoptosis and mitochondrial function. Results H2 02 in- duced intracellular ROS increased significantly apoptosis and necrosis cells, proanthocyanidins and blueberry proanthocyanidins extract and H2 02 combined treatment, intracellular ROS decreased, the percentage of necrotic cells has decreased. Conclusion Blueberry proanthocyanidins have a certain antioxidant activity, and the cytotoxicity is weak.
