摘  要:目的 为了探讨拟除虫菊酯对中枢神经系统损害的机理。方法 用逆转录 聚合酶链反应 (RT PCR) ,斑点杂交 ,流式细胞术及免疫组织化学技术同时检测了氯菊酯 (PM )和溴氰菊酯 (DM )对大鼠脑组织中神经生长因子 (NGF)表达的影响。结果4种方法检测结果均表明 1次大剂量 (PM1组 ,4 0 0mg·kg- 1)和连续 5d较小剂量 (PM2组 ,2 0 0mg·kg- 1)PM处理后大鼠脑组织NGF的mRNA和蛋白表达增多。其中RT PCR和斑点杂交结果表明各组大鼠大脑皮层和海马的NGFmRNA水平均有显著升高 ;其蛋白表达水平的变化与mRNA升高基本一致。而DM对NGF的影响与PM稍有不同 ,虽然大剂量(DM1组 ,2 5 .0mg·kg- 1) 1次ip和连续 5d较小剂量 (DM2组 ,12 .5mg·kg- 1)ip均可引起NGFmRNA表达的增加 ,流式细胞术结果却表明DM2组海马的NGF蛋白表达水平无明显的增加 ;免疫组化亦表明DM2组皮层及海马各区NGF的蛋白表达均无明显升高。结论 两型拟除虫菊酯对NGF蛋白表达的诱导作用不同 。AIM To investigate the mechanism of permethrin(PM) and deltamethrin(DM) on central nervous system. METHODS The levels of nerve growth factor(NGF) mRNA and protein expression in rat cortex and hippocampus were observed by reverse transcription-polymerse chain reaction(RT-PCR), dot blotting, flow cytometry analysis and immunohistochemistry. RESULTS The levels of NGF mRNA expression are up-regulated by PM administered at a higher dose (PM1 group, 400 mg·kg -1, ip) one time and a lower dose (PM2 group, 200 mg·kg -1·d -1, ip) daily for 5 d, and PM produced an increase in NGF protein expression in cortex and hippocampus showing a similar trend. However, another pyrethroid, DM, can not up-regulate NGF protein expression after continuous administration(DM2 group, 12.5 mg·kg -1·d -1, ip, for 5 d), the level of NGF mRNA was elevated at DM1 (25.0 mg·kg -1, ip, one time) and DM2 groups. CONCLUSION There is some difference between PM and DM in the ability in induction of NGF protein expression. Elevations of NGF expression in rat brain maybe a neuroprotective profiles in injury of central nervous system.
