摘 要:目的:观察不同压力灌注下上尿道梗阻大鼠肾损伤情况。方法:将80只SD大鼠随机分为8组,每组10只。M0~M3组制备轻度上尿路梗阻模型并分别给予0、20、60、100 mm Hg压力的肾盂灌注,S0~S3组制备重度上尿路梗阻模型并分别给予相应压力的肾盂灌注。灌注8 min后停止2 min,重复操作4次。术后48 h收集梗阻侧肾组织,检测肾小管上皮细胞凋亡指数,Western blot法检测肾损伤分子1(KIM-1)和富半胱氨酸蛋白61(Cyr61)的表达。结果:梗阻程度越重,灌注压力越高,肾小管上皮细胞凋亡指数越大,KIM-1和Cyr61的表达越强(P<0.05)。结论:上尿路梗阻降低了导致肾损伤的肾盂灌注压力阈值。Aim: To observe the degree of acute nephron injury in rats with upper urinary tract obstruction and different pressure perfusion. Methods: A total of 80 SD rats were equally divided into 8 groups( n = 10). The rats in groups M0,M1,M2 and M3 received an operation to induce mild hydronephrosis model and then were perfused with 0,20,60,and 100 mm Hg pressure fluid,respectively. The rats in groups S0,S1,S2 and S3 were made severe hydronephrosis model and then were perfused with corresponding pressure fluid. The operation was 8 min-perfusion and then 2 min-intermittent,and repeated 4 times. 48 h after the perfusion,the possible change of pathology in kidney tissue was investigated,the apoptosis index was detected,and the expression levels of kidney injury molecule 1( KIM-1)protein and cysteine-rich protein 61( Cyr61) were detected by Western blot. Results: The apoptosis index and the expression levels of KIM-1 and Cyr61 increased with the increase of hydronephrosis degree and perfusion pressure( P〈0. 05). Conclusion: Upper urinary tract obstruction may reduce the threshold of kidney perfusion pressure which could induce acute kidney injury.
