不同浓度DAPT对结核病患者PBMCs凋亡和增殖的影响

摘　　要：目的探讨γ-分泌酶抑制剂中的DAPT对结核病(TB)患者外周血单个核细胞(PBMCs)的凋亡和增殖作用。方法收集TB患者、潜伏感染者(LTBI)和结核菌素皮试实验阴性(TST-)健康者外周血标本各30例,淋巴细胞分离液分离PBMCs,应用不同浓度DAPT(0、2.5、5、10和20μmol/L)处理PBMCs,应用显微镜计数PBMCs数量,流式细胞术法测定细胞凋亡和细胞增殖情况。结果与DMSO组比较,TST-和LTBI组细胞数均随着DAPT浓度增加剂量依赖性降低,而TB患者在DAPT为10μmol/L时细胞数量减少明显,凋亡明显增加,差异有统计学意义(P<0.05),在20μmol/L DAPT时细胞数量明显增加,凋亡降低。但是,与DMSO组比较,每组不同浓度DAPT组细胞增殖,差异无统计学意义(P>0.05)。结论 DAPT可以增加结核病患者PBMCs凋亡,在DAPT浓度为10μmol/L时凋亡最明显,但不影响细胞增殖。Objective To study the apoptotic and proliferation effect of gamma-secretase inhibitor（DAPT）on peripheralblood mononuclear cells（PBMCs） of patients with tuberculosis（TB）. Methods Peripheral blood specimens werecollected from TB patients,latent tuberculous infection（LTBI）and tuberculin skin test negative（TST-）individuals;eachgroup had 30 cases. PBMCs were separated by ficoll-hypaque solution;PBMCs were treated by different concentrations（0,2.5,5,10 and 20 μmol/L）of DAPT;the number of PBMCs was counted by microscope;cell apoptosis and proliferationwere determined by flow cytometry method. Results Compared with DMSO group,PBMCs number was reduced in a dosedependent in TST negative and LTBI groups,while when DAPT was 10 μmol/L,the PBM cells were decreased significantlyand apoptotic cells were obvious in TB patients（P〈0.05）. However,PBM cells increased significantly in 20 μmol/L DAPT,and apoptotic cell decreased. But compared with DMSO group,cell proliferation was not affected in all three groups（P〈0.05）. Conclusion DAPT could increase cell apoptosis at 10 μmol/L DAPT,but not affect proliferation in TB patients.
