摘  要:为探明miR-450基因家族(miR-450s)的系统进化历程及其在沙子岭猪睾丸组织不同发育时期表达规律。通过miRBase数据库检索miR-450s序列,利用Ensembl数据库信息确定miR-450s在不同物种中的基因组定位,采用MEGA 6.0构建miR-450基因家族的系统进化树,并利用RT-q PCR检测miR-450s在沙子岭猪睾丸组织不同发育期(E90、D1、D30、D60、D90、D120、D150、D180)的表达变化。miRBase数据库共检索15个物种36条miR-450s序列,且miR-450s序列具有高度保守性。基因组定位显示miR-450s位于X染色体上,均位于基因间隔区(Intergenic region,IGR),且miR-450基因家族成员在进化过程中紧密相连,可能由于串联重复所致。RT-q PCR结果显示,miR-450s在胚胎期中表达量显著高于出生后表达量(P<0.01),且miR-450基因家族3个成员(miR-450a、miR-450b、miR-450c)在沙子岭猪睾丸不同发育阶段呈现出协同表达的特性。miR-450基因家族可能在猪睾丸组织发育过程中发挥重要调控作用。This experiment was conducted to investigate the evolutionary process of miR-450 gene family (miR-450s) and its expression pattern in the testis of Shaziling pig. The sequences of miR- 450 gene family members from different animal species were searched from rniRBase database, the miR-450 genes were located by Ensembl database, its phylogenetic tree was constructed by MEGA 6.0, and the expression levels of miR-450s were detected in different developmental stages (E90, D1, D30, D60, D90, D120, D150, D180) of porcine testieular tissue by real-time fluorescence quantitative PCR. A total of 36 sequences from 15 species were searched and all members of the miR-450 gene family were highly conserved. The analysis of localization showed miR-450s were loca- ted on the intergenic region (IGR) of X chromosome, and miR-450s gene were closely linked in the evolutionary process, which maybe caused by tandem repeats. RT-qPCR results showed that the ex- pression levels of miR-450s gene in the embryonic period were extremely significant higher than that in postnatal periods (P〈0. 01), and the 3 members (miR-450a, miR-450b, miR-450c) of miR- 450 gene family showed the characteristics of co-expression in different development stages, miR-450 gene family may play an important role in the development of the pig testicular tissue.
