含SLH的苏云金芽胞杆菌菌株的筛选

摘　　要：本实验通过采集河边、污水池边、草坪上和花坛中的土壤样本,利用平板划线法分离和纯化目标菌株,获得菌落形态类似于苏云金芽胞杆菌菌株。再通过相差镜检观察其是否形成透明的芽胞及致密的深色的伴胞晶体,以及利用SDS-PAGE检测晶体蛋白的表达来筛选苏云金芽胞杆菌。提取菌种的基因组DNA并进行PCR,扩增S-层蛋白特异性的保守SLH序列,获得的阳性菌株即为含SLH的苏云金芽胞杆菌,最后筛选到5株包含SLH的苏云金芽胞杆菌。This experiment mainly research contents for thuringiensis containing S - layer of bacillus coli strain screening. Through acquisition on the lake, sewage pool, lawn and soil samples from the flower bed, using the method of flat line, separation, purification and selection for colony morphology is similar to some 8 strains of bacillus coli strains. Whether through microscopy to observe the difference form transparent buds and the companion cells of some square or circular crystal and using sds-page detection of crystal protein expression. Strains of extraction of genomic DNA and PCR, the amplification of S - layer protein specificity conservative SLH series, the positive strains is the SLH - domain thuringiensis of bacillus coli.
