摘 要:目的:观察益气解毒活络方对链脲佐菌素(streptozotocin,STZ)诱导的糖尿病(diabetes mellitus,DM)大鼠胃组织中Bcl-2、Bax及促生长素(ghrelin)表达的影响。方法:高健康SPF级Wistar大鼠55只,随机分为正常组10只,造模组45只。造模成功后将大鼠随机分为模型组,中药组,西药组各10只,西药组以西沙比利(西安杨森)0.5 mg·(kg·d)^(-1)灌胃,中药组按人与大鼠等效剂量的6.3倍计算[12.78 g·(kg·d)^(-1)]灌胃。正常组与模型组给予等量生理盐水灌胃,每日1次,连续8周。实验期间大鼠自由饮水,不使用降糖药及胰岛素等药物。每周称量一次体质量,根据体质量调整灌胃量。实验结束后,检测各组大鼠体质量(body weight,BW)、空腹血糖(fasting blood-glucose,FBG)水平;免疫组化法观察大鼠胃组织中Bcl-2、Bax蛋白表达,实时荧光定量PCR法检测胃窦组织中ghrelin mRNA表达。结果:与正常组比较,模型组大鼠体质量明显降低,FBG明显升高,胃组织中Bcl-2蛋白表达及ghrelin mRNA表达降低,Bax蛋白表达升高(P<0.01);与模型组比较,各治疗组大鼠体质量增加,FBG明显降低,各治疗组大鼠上述其他情况明显改善(P<0.01)。结论:益气解毒活络方可以改善DGP大鼠胃组织Bcl-2、Bax及促生长素的表达,保护胃功能。Objective: To observe the effect of Yiqi Jiedu Huoluo Prescription on the expression of Bcl-2,Bax and somatotropin in the gastric tissue of DM rats induced by STZ. Methods: Totally 55 Wistar rats of SPF grade were randomly divided into normal group(10 cases) and modeling group(45 cases). After modeling,the rats were randomized into model control group,Chinese medicine treatment group and western medicine control group,10 cases in each group. Western medicine group were given gavage of Cisapride of 0. 5 mg·kg^(-1)·d^(-1). Chinese medicine group were given gavage with equivalent dose of 12. 78 g·kg^(-1)·d^(-1).Normal group and model group were given same amount of normal saline(NS) for gavage,once a day and for continuous8 weeks. During experiment,rats drunk water freely and weren' t given hypoglycemic drugs and insulin. The body weight were weighed once every week and regulate the gavage amount according to body weight. After experiment,the levels of BW and FBG were measured. The expression of Bcl-2 and Bax were observed by immunohistochemistry. The expression of ghrelin mRNA in the antrum of stomach was detected by PCR. Results: Compared with normal group,the body weight in model groups declined remarkably while the level of FBG increased obviously. The expression of Bcl-2 and ghrelin mRNA in the gastric tissue declined while the expression of Bax increased(P 0. 01). Compare with the model group,the BW increased and the FBG decreased in treatment group. The above conditions of rats in each group improved significantly(P 0. 01). Conclusion: The change of the expression of Bcl-2,Bax and ghrelin in the gastric tissues of DM rats can induce apoptosis of the gastric cells,which may be one pathogenesis of DGP. Yiqi Jiedu Huoluo Prescription can exert protective effect on the stomach through above scheme.
