摘 要:目的:观察解郁胶囊(蒲郁胶囊)对药物所致抑郁模型小鼠的影响及可能作用机制。方法:小鼠根据体质量随机分成空白组、丙咪嗪组、解郁胶囊(蒲郁胶囊)低剂量组(235 mg·kg^(-1))、解郁胶囊(蒲郁胶囊)中剂量组(470 mg·kg^(-1))、解郁胶囊(蒲郁胶囊)高剂量组(940 mg·kg^(-1))。通过利血平拮抗实验制造利血平抑郁模型,并观察解郁胶囊(蒲郁胶囊)对利血平所致的体温下降、运动不能、眼睑下垂的影响;通过五羟色氨酸诱导的甩头实验和育亨宾毒性实验观察小鼠甩头次数和育亨宾致死率,观察药物对小鼠体内五羟色胺和去甲肾上腺素的影响。结果:注射利血平后动物出现利血平化,即体温下降、运动不能及眼睑下垂,解郁胶囊(蒲郁胶囊)各剂量均可明显对抗其作用;在五羟色氨酸诱导小鼠甩头实验中观察到解郁胶囊(蒲郁胶囊)小鼠甩头反应次数未明显增加;在育亨宾毒性实验中观察到中剂量、高剂量解郁胶囊(蒲郁胶囊)均能增加育亨宾毒性。结论:解郁胶囊(蒲郁胶囊)在利血平诱导的小鼠抑郁模型上具有抗抑郁作用,其作用机制可能与增强脑内去甲肾上腺素能神经功能有关。Objective:To observe the impacts of Jieyu capsule(Puyu capsule) on mice of drug-induced depression model and the possible mechanism of action.Methods:The mice were divided randomly into control group,imipramine group,Jieyu capsule(Puyu capsule) low-dosage group(235 mg·kg^(-1)),Jieyu capsule(Puyu capsule) medium-dosage group(470 mg·kg^(-1)) and Jieyu capsule(Puyu capsule) high-dosage group(940 mg·kg^(-1)) according to body weights.A reserpine-induced depression model was produced through reserpine antagonism experiment,and the impacts of Jieyu capsule(Puyu capsule) on reserpine-induced body temperature drop,acinesia and ptosis were observed;then the times of mice twitching heads and fatality rate caused by yohimbine were observed through serotonine-induced head twitching experiment and yohimbine toxicity experiment,and also the impacts of drugs on serotonine and norepinephrine in mouse body were observed.Results:After being injected with reserpine,animals were reserpinized,i.e.,subjected to body temperature drop,acinesia and ptosis;Jieyu capsule of different dosages could antagonize its actions remarkably.In serotonine-induced head twitching experiment,it was observed that the times of head twitching of the mice taking Jieyu capsule(Puyu capsule) hadn't increased remarkably.In yohimbine toxicity experiment,it was observed that the mediumdosage and high-dosage Jieyu capsule(Puyu capsule) could both add yohimbine' s toxicity.Conclusion:Jieyu capsule(Puyu capsule) has the anti-depression effect on reserpine-induced depression model mice;its mechanism of action is likely to be related to the enhancement of neurologic function of norepinephrine in the brain.
