摘 要:目的探讨氧化钕颗粒物对大鼠肺泡巨噬细胞NR8383细胞毒性作用及氧化损伤作用。方法将处于对数生长期的NR8383细胞,暴露于终浓度为0、3.125、6.25、12.5、25和50μg/ml氧化钕混悬液6、12和24 h后,采用MTT法检测NR8383细胞抑制率;终浓度为0、3.125、6.25、12.5、25和50μg/ml氧化钕混悬液暴露24 h后,测定细胞培养液上清中乳酸脱氢酶(lactate dehydrogenase,LDH)、总抗氧化能力(total antioxidant capacity,T-AOC)、超氧化物歧化酶(superoxide dismutase,SOD)和丙二醛(malondialdehyde,MDA)的浓度。结果与对照组比较,各浓度氧化钕暴露不同时间后NR8383细胞的抑制率均较高,差异有统计学意义(P<0.05);与对照组比较,稀土氧化钕暴露NR8383细胞24 h后,12.5、25和50μg/ml染毒组随着染毒剂量增加,LDH水平逐渐升高;50μg/ml染毒组的T-AOC水平和12.5、25和50μg/ml染毒组的SOD水平低于对照组,25和50μg/ml染毒组MDA水平高于对照组,差异有统计学意义(P<0.05)。结论氧化钕颗粒物暴露可致细胞毒性作用和氧化损伤作用。Objective To investigate the toxicity and oxidative damage of neodymium oxide particles in pulmonary macrophage NR8383 cells. Methods NR8383 cells in the logarithmic growth phase were exposured to neodymium oxide suspension with final concentration of 0,3. 125,6. 25,12. 5,25,50 μg/ml for 6,12,24 h. MTT method was used to assay the cell inhibition rate. After exposure to final concentration of 0,3. 125,6. 25,12. 5,25,50 μg/ml of neodymium oxidesuspension for 24 h,the content of lactate dehydrogenase,(lactate dehydrogenase,LDH),total antioxidant capacity(T-AOC),superoxide dismutase(SOD) and malondialdehyde(MDA) of NR8383 cells in the logarithmic growth phase were measured. Results Compared with the control group,the inhibition rate of NR8383 were increased after exposure to neodymium oxide at different concentration and time. The difference was statistically significant(P〈0. 05).Compared with the control group,the concentration of LDH in NR8383 cell increased with the increasing concentration of neodymium oxide which was 12. 5,25,50 μg/ml. The levels of T-AOC and SOD were lower than that in control group,and the content of MDA was higher than that in control group,the above-mentioned difference were significant(P〈0. 05); Conclusion Neodymium oxide particles might induce cell toxicity and oxidative damage.
