摘 要:目的观察石斛合剂序贯法对糖尿病合并肝纤维化大鼠肝功能及肝脏形态学的影响。方法采用高脂高糖联合小剂量STZ建立2型糖尿病模型,再皮下注射CCl4建立糖尿病合并肝纤维化的动物模型,尾静脉采血测血糖;紫外比色法测定AST、ALT;HE染色及Masson染色观察肝形态;ELISA检测肝纤维化四项。结果与干预前比较,干预后二甲双胍组和石斛合剂组血糖明显下降(P<0.05);与模型对照组相比,二甲双胍组和石斛合剂组血糖、AST、ALT、肝纤维化四项指标均明显降低(P<0.05);肝脏HE染色和Masson染色均显示二甲双胍组和石斛合剂组炎性浸润减少,汇管区及中央静脉区胶原纤维沉积较模型对照组减轻,且石斛合剂组较二甲双胍组效果更显著。结论石斛合剂序贯法能显著抑制肝纤维化的进展,且明显优于单纯用二甲双胍治疗。Objective: To observe the effect of DMOC on liver function and liver morphology in diabetic rats with hepatic fibrosis. Method: The model of type 2 diabetes mellitus was established by high fat and high glucose combined with low dose of STZ. Animal models of diabetic and hepatic fibrosis were established by injecting CCl4 subcutaneously. Blood was collected from the tail vein for blood glucose measurement. AST and ALT were determined by UV colorimetry. HE staining and Masson staining Observed liver morphology; ELISA measure Liver fibrosis indicators. Results: Compared with those before intervention, the blood glucose of metformin group and DMOC group were significantly decreased after intervention(P〈0.05). Compared with the model control group, the blood glucose, AST, ALT and liver fibrosis were significantly increased in metformin group and DMOC group(P〈0.05). HE staining and Masson staining showed that the inflammatory infiltration of metformin group and DMOC group were decreased, the deposition of collagen fibers in the portal area and the central venous area was relieved compared with the model group, and the DMOC group was more effective than metformin group. Conclusion: DMOC can significantly inhibit the progress of liver fibrosis, and was significantly better than the simple treatment with metformin.
