摘 要:目的通过比较流感病毒感染小鼠与正常小鼠、达菲干预组血清之间的拉曼光谱差异,探索基于表面增强拉曼光谱的流感病毒感染血清检测新方法。方法以纳米银溶胶为活性基底,采用便携式拉曼光谱仪分别检测正常组、模型组和达菲组小鼠血清的拉曼增强光谱,并进行正交校正的偏最小二乘辨别分析(OPLS-DA)。同时,采用RT-PCR测定肺组织中流感病毒RNA复制数作为对照方法。结果在第3、5天两个时间点,正常组、模型组和达菲组3组血清均表现出较为明显的分离趋势,经ROC曲线评估,运用OPLS-DA建立的3组血清SERS图谱模型预测能力较高,能够对3组血清进行良好的区分、鉴别。结论 SERS与RT-PCR检测鉴别结果一致,初步研究表明SERS图谱有助于鉴别和诊断机体是否感染了流感病毒。Objective A new method for serum detection based on surface-enhanced Raman spectroscopy was explored by comparing the Raman spectra between normal mice and influenza virus-infected mice. Methods The nano-silver sol was used as the active substrate. The Raman spectra of the normal group, the model group and the Tamiflu control group were detected by portable Raman spectroscopy, and the partial least squares discrimina- tion analysis (OPLS-DA) was performed by the orthogonal correction. The number of RNA replicas of influenza virus in lung tissue was measured by RT-PCR as a control method. Results At the 3rd or 5th days, the serum of the normal group, the model group and the Tamiflu control group showed a significant trend. By the ROC curve evaluation, the predictive ability of 3 groups of serum SERS models established by OPLS-DA was high, which could distinguish and differentiate 3 groups of serum. Conclusions The results of SERS and RT-PCR detections were consistent. The preliminary results show that SERS pattern can help to identify and diagnose whether the body is infected with influenza virus.
