摘 要:目的通过研究富血小板血浆(PRP)联合转化生长因子(TGF)-β1抗体对骨骼肌损伤后修复过程的作用,探索减少纤维化和促进骨骼肌再生的方法。方法选择雄性SD大鼠30只,鼠龄6个月,体质量约250 g。随机分为PRP抗体组(A组)、PRP模型组(B组)和模型对照组(C组),每组10只。3组全部采用0.5%布比卡因溶液注射大鼠胫骨前肌制作骨骼肌损伤模型,造模后第2天,A组于损伤处注射PRP联合TGF-β1抗体,B组单独注射PRP,C组注射等量0.9%氯化钠溶液(生理盐水)。各组大鼠于实验第14天全部处死、取材,进行Masson染色,观察组织形态学变化;免疫组织化学染色观察TGF-β1、PAX-7表达情况。结果在Masson染色中发现,3组损伤骨骼肌都出现纤维化改变,但是A组比B组和C组更少的表达纤维化程度(P<0.01)。免疫组织化学染色观察到A组TGF-β1表达水平[灰度值(0.13±0.01)]也比B组(0.15±0.02)和C组(0.19±0.02)明显减少(P<0.05),而A组PAX-7表达水平[灰度值(0.25±0.02)]则比B组(0.22±0.03)和C组(0.19±0.02)明显增加(P<0.05)。结论 PRP联合TGF-β1抗体可以明显抑制骨骼肌损伤后胶原纤维的表达,同时增强骨骼肌卫星细胞活化和增殖,更好促进损伤骨骼肌修复。Objective To study the repair effects of platelet-rich plasma(PRP) combined with transforming growth factor-β1 antibody on skeletal muscle injury in rats, and explore the method of reducing fibrosis and promoting skeletal muscle regeneration. Methods Thirty male rats of 6-month old with body weight of 250 g were randomly divided into injury PRP antibody group(group A), PRP model group(group B) and model control group(group C), 10 in each group. All of the rats were made skeletal muscle injury model by injection of 0.5 % bupivacine in tibialis anterior muscle. On second day after modeling, group A received PRP combined with TGF-β1 antibody injection on the injury, group B received only PRP, while group C received the same amount of 0.9 % sodium chloride solution(saline). All rats were sacrificed on 14 th day after experiment, and tibialis anterior muscles were removed for histological analysis. The histological changes were observed with Masson staining and im-munohistochemical staining was used to observe the expression of TGF-β1 and PAX-7. Results The Masson staining results showed that fibrosis changes of injured skeletal muscle were observed in 3 groups, with group A expressed less fibrosis than group B and C(P〈0.01). The immunohistochemical staining results showed that the expression of TGF-β1 in group A[grayscale value(0.13 ± 0.01)] was significantly lower than that in group B[grayscale value(0.15 ± 0.02)] and group C[grayscale value(0.19 ± 0.02)](P〈0.05), while the expression level of PAX-7 in group A[grayscale value(0.25 ± 0.02)] significantly higher than that in group B[grayscale value(0.22 ± 0.03)] and group C[grayscale value(0.19 ± 0.02)](P〈0.05). Conclusion It is demonstrated that PRP combined with TGF-β1 antibody will significantly decrease the fibrosis and prolong satellite cell activation,which promote the regeneration after muscle injury.
