摘 要:目的:研究吴茱萸碱对动脉粥样硬化(AS)大鼠模型血清中ox-LDL及炎性小体caspase-1表达的影响,从抗炎抗氧化作用方面探讨吴茱萸碱抗动脉粥样硬化作用及机制。方法:将60只SD大鼠随机分为6组,即空白组、模型组、辛伐他汀对照组和吴茱萸碱高、中、低剂量组,每组10只。适应性喂养1周后,以高脂饲料喂养4周造模。造模成功后,分别给予辛伐他汀、吴茱萸碱(高、中、低剂量)灌胃治疗,3周后处死大鼠,检测血清中TG、TC、HDL、LDL含量及ox-LDL、caspase-1的表达量;观察HE染色切片大鼠主动脉弓粥样硬化斑块及肝细胞结构变化。结果:HE染色见大鼠肝组织及主动脉弓组织中吴茱萸碱给药组炎症细胞浸润及脂肪变性较模型组明显减轻,其中吴茱萸碱中剂量组效果最为显著;与模型组比较,吴茱萸碱能显著提高AS大鼠模型HDL血清中的含量,显著降低TC、TG、LDL水平及ox-LDL、caspase-1含量,其中吴茱萸碱中剂量效果最为明显(P<0.05或P<0.01)。结论:吴茱萸碱可通过抗炎抗氧化途径发挥抗AS作用。Objective:To study the effect of Evodiamine on the expression of ox-LDL and inflammatory body caspase- 1 in serum of atherosclerosis (AS) rats, and to discuss the anti-atherosclerosis effect of Evodia- mine and its mechanism in terms of anti-inflammation and anti-oxidation. Methods : A total of 60 SD rats were randomly and evenly divided into 6 groups: a blank group, a model group, a Simvastatin control group, and three groups of Evodiamine with high, middle and low dose. After one week of adaptive feeding, the model was made by feeding with high fat diet for 4 weeks. After successful modeling, Simvastatin, Evodiamine ( with high, middle and low dose ) were given to lavage. Three weeks later, the rats were killed to detect the contents of TG, TC, HDL, LDL in serum and the expression of ox-LDL and caspase-1. The changes of Atherosclerotic plaque and Hepatocyte structure of Aortic Arch in Rats stained with HE were observed. Results: HE staining showed that the infiltration of inflammatory cells and steatosis in the Evodipine administration group was signifi- cantly less than that in the model group, especially in the the middle-dose group. Compared with the model group, Evodiamine could significantly increase the content of HDL in serum of AS rats, and significantly de- crease TC, TG, LDL and ox-LDL and the content of Caspase- 1. The effect was the greatest in the middle-dose group (p〈 0.05 or p〈0.01). Conclusion: Evodiamine can play the role of anti-AS by anti-inflammatory and anti -oxidant.
