摘 要:目的应用生物信息学方法分析人H10N8禽流感病毒血凝素蛋白(hemagglutinin,HA)的主要特性,预测可能的B/T细胞抗原表位。方法采用Prot Param预测H10N8禽流感病毒HA蛋白的理化特性,SOPMA预测二级结构,Motif Scan预测翻译后修饰位点,DNAStar分析其序列的亲水性指数、柔韧性指数、可及性参数以及抗原指数,推测B细胞抗原表位的可能位置,并采用SYFPEITHI的T细胞表位预测工具分别预测CTL和Th细胞表位。结果人易感H10N8禽流感病毒HA为亲水性蛋白,二级结构中包含218个α-螺旋、108个β-片层、64个β-转角和171个无规卷曲,具有多个糖基化、酰胺化及磷酸化翻译后修饰位点。经综合各种参数,预测出H10N8禽流感病毒HA蛋白可能存在的7个B细胞抗原表位、7个CTL细胞表位和8个Th细胞表位。结论 HA蛋白B/T细胞抗原表位的预测为人易感H10N8染禽流感病毒的疫苗研制奠定了一定的基础。Objective To predict the main characteristics and B/T cell epitopes of hemagglutinin( HA) of human-infecting avian influenza A( H10 N8). Methods The physical and chemical properties were predicted by using Prot Param,and secondary structure was predicted by using SOPMA online server. The post translation sites were predicted by using Motif Scan software. The B cell epitopes were analyzed by DNAStar software,which can analyze the hydrophilicity,flexible regions,surface accessibility regions and antigenic index. The T cell epitopes were analyzed by SYFPEITHI. Results HA of influenza A( H10 N8) was a hydrophilic protein,which had 218 α-helixs,108 β-sheets,64 β-turns and 171 random coils,and also had some post-translational modification sites. In addition,7 potential B cell epitopes,7 potential CTL cell epitopes and 8 potential Th cell epitopes of HA protein were predicted. Conclusion The B/T cell epitopes prediction might be useful to develop some promising anti-H10 N8 vaccines based on the peptide.
