摘 要:目的观察鱼腥草水提液及其主要成分绿原酸、芦丁、金丝桃苷、槲皮素体外抗肠道病毒71型(EV71)的作用。方法将RD细胞(来源于人横纹肌肉瘤细胞)分为鱼腥草组、金丝桃苷组、槲皮素组、芦丁组、绿原酸组、对照组。鱼腥草组分别给予5 000、2 500、1 250、625、313 mg/L的鱼腥草水提液;槲皮素组加入槲皮素药液,芦丁组加入芦丁药液,药液浓度分别为10.00、5.00、2.50、1.25、0.63 mg/m L;金丝桃苷组加入金丝桃苷药液,绿原酸组加入绿原酸药液,药液浓度分别为4.00、2.00、1.00、0.50、0.25 mg/m L;对照组加入细胞培养液。在EV71感染细胞的不同阶段各组加入相应药液,观察细胞病变(CPE),分别计算病毒复制抑制率、半数抑制浓度(IC50)、病毒生长阻滞率、病毒吸附抑制率、病毒灭活率、病毒穿入细胞抑制率。结果鱼腥草组、槲皮素组病毒复制抑制率高于其余四组,且药物作用浓度越高,抑制作用越强(P均<0.05);金丝桃苷组和芦丁组病毒生长阻滞抑制率高于其余四组(P均<0.05);金丝桃苷组、鱼腥草组病毒吸附抑制率高于其余四组(P均<0.05)。鱼腥草水提液及其主要成分均没有灭活病毒的作用。各组在作用15、30 min时均未表现出对病毒穿入的抑制作用;在作用60 min时,鱼腥草组、槲皮素组、芦丁组、绿原酸组病毒穿入抑制率高于金丝桃苷组和对照组(P均<0.05);槲皮素组和芦丁组病毒穿入抑制率随药物浓度降低而减弱,绿原酸组病毒穿入抑制率随药物浓度降低而增强。结论鱼腥草水提液及主要成分都具有一定的体外抗EV71作用,但无灭活病毒的作用。其中,槲皮素具有抑制病毒复制的作用,金丝桃苷和芦丁有阻滞病毒生长作用,金丝桃苷还可抑制病毒吸附,槲皮素、芦丁、绿原酸有抑制病毒穿入的作用。Objective To observe the anti-enterovirus type 71( EV71) effects of Houttuynia cordata Thunb water extract and its main ingredients,including chlorogenic acid,rutin,spasinin,and quercetin in vitro. Methods The RD cells( derived from human rhabdomyosarcoma cells) were divided into the Houttuynia cordata group,hyperoside group,quercetin group,rutin group,chlorogenic acid group,and control group. Cells in the Houttuyniae cordata group were given water extracts of Houttuynia cordata at the concentrations of 5 000,2 500,1 250,625,and 313 mg/L; cells in the quercetin and rutin groups were added with quercetin and rutin at the concentrations of 10. 00,5. 00,2. 50,1. 25,and 0. 63 mg/m L;cells in the hyperoside and chlorogenic acid groups were given hyperoside and chlorogenic acid,respectively,with the concentrations of 4. 00,2. 00,1. 00,0. 50,and 0. 25 mg/m L; cells in the control group were added with the cell culture fluid. We observed the cytopathic effect( CPE) and calculated the inhibition rate of virus replication,half maximal inhibitory concentration( IC50),inhibition rate of virus growth,inhibition rate of virus adsorption,virus inactivation rate,and virus penetration rate. Results The inhibition rate of virus replication in the Houttuynia cordata group and quercetin group was higher than that in the other four groups,and the inhibition rate increased with the increasing concentrations( all P 0. 05). The inhibition rate of virus growth in the hyperin and rutin groups was higher than that in the other four groups( all P〈 0. 05). The inhibition rate of virus adsorption in the hyperin and Houttuynia cordata group was higher than that in the other four groups( all P〈 0. 05). The water extract and its main components of Houttuynia cordata had no effect in inactivating the virus. No inhibitory effect on virus penetration was observed in each group at 15 and 30 min.
