ENTRESTO对心力衰竭大鼠血清cAMP、BNP、Gal-3及生存时间的影响

摘　　要：目的研究ENTRESTO对心力衰竭大鼠血清c AMP、BNP、Gal-3及生存时间的影响。方法将SD雄性大鼠采用结扎冠状动脉左前降支近端的方法建立心肌梗死后心力衰竭大鼠模型,将其随机分为4组:正常组、建模组及低、高剂量实验组(sacubitril/缬沙坦,49/51 mg,97/103 mg),每组8只,灌胃给药。正常组及建模组给予同等剂量蒸馏水。分别于给药后3、9、18 h取大鼠静脉血,采用化学发光法测定BNP;采用ELISA检测法测定c AMP、Gal-3;观察小鼠生存时间。结果建模组大鼠个时段血清Gal-3、BNP含量较其他组最高,差异具有统计学意义(P<0.05),经ENTRESTO治疗后,低、高剂量组大鼠血清Gal-3、BNP水平均有不同程度下降,且高剂量组大鼠Gal-3、BNP水平低于低剂量组,高于正常组(P<0.05)。建模组大鼠个时段血清c AMP含量较其他组最低,差异差异具有统计学意义(P<0.05),经ENTREST治疗后,低、高剂量组大鼠血清c AMP水平均有不同程度升高,且高剂量组大鼠c AMP水平高于低剂量组,低于正常组,差异具有统计学意义(P<0.05)。建模组大鼠最早出现死亡个体,且最早全部死亡;经生存分析得知,与低、高剂量组对比,差异显著,具有统计学差异(P<0.05)。结论ENTRESTO能够降低心力衰竭大鼠血清中的Gal-3、BNP水平,升高c AMP水平,延长心力衰竭大鼠生存时间。Objective To study the effects of ENTRESTO on serum cAMP, BNP, Gal-3 and survival time in rats with heart failure. Methods SD male rats with heart failure by ligating the left anterior descending branch proximal which were established after myocardial infarction, were randomly divided into 4 groups：normal group,model group, low and high dose experimental groups （sacubitril/Valsartan, 49/51 mg, 97/103 mg）, with 8 rats in each group, and conducted intragastric administration. The normal group and the model group were given the same amount of distilled water. Venous blood of rats was taken at 3,9 and 18 h after administration, and BNP was detected by using chemiluminescence. CAMP and Gal-3 were detected by ELISA assay, and the survival time was measured. Results The model group rats hadthe highest serum Gal- 3, BNP content than other groups （ P 〈 0.05 ）. After the ENTRESTO treatment, for both low dose and high dose groups, the rats serum Gal-3, BNP levels were decreased, and the Gal-3 and BNP levels of rats in high dose group was lower than in low dose group, and higher than the normal group （ P 〈 0.05 ）. For modeling group, the rats serum cAMP content was lower than other groups （ P 〈 0.05 ）. For low dose and high dose groups, the rat serum cAMP levels were both elevated, and the level of cAMP in high dose group were higher than that of low dose group, lower than the normal group （ P 〈 0.05 ）. The rats in the modeling group died first and died for all at the earliest point. The survival analysis showed that the difference between this group and the low and high does groups were statistically significant（P 〈 0.05）. Conclusion ENTRESTO can reduce the levels of Gal-3 and BNP in the serum of rats with heart failure,and increase the level of cAMP, and prolong the survival time of CHF rats.
