MTT法检测夏枯消瘤丸含药血清对人肺癌A549细胞的实验研究

摘　　要：目的:通过体外实验观察夏枯消瘤丸含药血清对人肺癌A549细胞的细胞毒作用。方法:MTT比色法检测人肺癌A549细胞的增殖,探讨夏枯消瘤丸含药血清对人肺癌A549细胞的细胞毒作用,从而明确夏枯消瘤丸对肿瘤的抑制作用;观察各组对人肺癌A549细胞的细胞形态及生长状态的影响,从而探究抑制肿瘤生长的有效浓度及剂量。结果:与正常对照组相比,5%含药血清组24 h细胞抑制率差异无统计学意义(P>0.05),其余含药血清组、DDP及综合组抑制率升高(P<0.05);随着时间延长,48 h、72 h抑制率也随之升高(P<0.05)。结论:夏枯消瘤丸含药血清对A549细胞增殖有明显抑制作用,并随血药浓度增大抑制作用增强,但不及DDP单用。Objective：To observe the cytotoxic effect of Xiakuxiaoliu pill containing serum on human lung cancer A549 cells in vitro. Mcthods：MTT assay was used to detect the proliferation of human lung cancer cell line A549 and to explore the cytotoxic effect of the herb-containing serum of Xiakuxiaoliu pill on human lung cancer cell line A549 so as to clarify the inhibitory effect on tumor cells. The effects of different groups and herb-containing serum concentration on the cell morphology and growth of human lung cancer A549 cells were observed to explore the effective concentration and dose of xiakuxiaoliu pill on inhibiting tumor growth. Results：Compared with the normal control group, the inhibitory rate of 24 h in 5% serum group had no significant difference （P〉0.05）, and inhibition rate was increased in the other herb-containing serum group, DDP group and combined group（P〈0.05）. With the extension of time,the inhibitory rates were increased at 48 h and 72 h （P〈0.05）. Conclusion ：The herb-containing serum of Xiakuxiaoliu pill group have a significant inhibitory effect on the proliferation of A549 cells,and its inhibitory effect is enhanced with the increase of plasma concentration,but it is inferior to that of DDP alone.
