膝骨性关节炎患者关节液中miRNA-146a、MMP-13的表达分析及相关性研究

摘　　要：目的:检测膝骨性关节炎(KOA)患者与非(KOA)患者关节液中miR-146a和MMP-13的表达情况,并探讨miRNA家族链与该疾病的相关性。方法:采用荧光定量PCR技术检测miRNA-146a、MMP-13在KOA组(n=40)与非KOA组(n=40)关节液中的表达水平,并运用Spearman分析miRNA-146a与MMP-13的相关性。结果:两组患者关节液中的目的基因均能检测到,扩增曲线良好,扩增率高;KOA组患者膝关节液中的miRNA-146a表达量显著低于非KOA组,MMP-13表达量明显高于非KOA组,差异均有统计学意义(P<0.05);相关性分析显示,KOA组关节液中miRNA-146a和MMP-13之间呈明显负相关性(r=-0.675,P<0.05)。结论:miRNA-146a与MMP-13的负相关性表达与膝骨性关节炎的发病机制有相关性,且呈现高MMP-13、低miRNA-146a表达。Objective with knee osteoarthritis To investigate the expression of miR-146a and MMP-13 in synovial fluid of patients （KOA） and non-KOA patients, and to explore the correlation between miRNA family chain and the disease. Methods： The expression levels of miRNA-146a and MMP-13 in the synovial fluid of KOA group （n=40） and non-KOA group （n=40） were detected by fluorescence quantitative PCR. The relevance of miRNA-146a and MMP-13 was analyzed by Spearman eorrelation analysis. Results： （1） The target gene in synovial fluid of the two groups can be detected; the amplification curve is good, high expansion rate. （9） The expression of miRNA-146a in knee joint fluid of KOA group was signifieantly lower than that of non-KOA group, while the expression of MMP-13 was signifieantly higher in KOA group than the non-KOA group （P〈 0.05）. （3） Correlation analysis of the factors within the KOA group showed a significant negative correlation be- tween miRNA-146a and MMP-13 （r=-0.675, P〈0.05）. Conclusion： The negative correlation between miRNA- 146a and MMP-13 expression is related to the pathogenesis of knee osteoarthritis, and present the high MMP- 13 and low miRNA-146a expression.
