摘  要:目的:优化大孔树脂吸附法富集锦鹤养心方总黄酮的工艺条件,并研究其对小鼠常压耐缺氧的影响。方法:以芦丁为对照品,硝酸铝显色法检测总黄酮的含量,通过静态吸附/解吸与动态吸附/解吸相结合的方法筛选大孔树脂型号,采用单因素实验考察上样量、上样流速、洗脱流速等对总黄酮纯化工艺的影响;采用常压耐缺氧实验,观察锦鹤养心方总黄酮对小鼠耐缺氧能力的影响。结果:AB-8型大孔树脂对锦鹤养心方总黄酮的纯化效果最好,最佳纯化工艺为上样液质量浓度0.5 g·m L-1,上样流速为2 BV·h-1,上样量为2 BV,吸附4 h,加水1.5 BV洗脱水溶性杂质,加50%乙醇3 BV洗脱,洗脱流速为2 BV·h-1,在此条件下,提取物中总黄酮质量分数由25.01%提高到85.54%;与空白对照组相比,锦鹤养心方总黄酮能明显增加小鼠常压耐缺氧存活时间。结论:AB-8型大孔树脂对锦鹤养心方总黄酮具有良好的纯化效果,优选工艺合理、稳定可行,且锦鹤养心方总黄酮能显著提高小鼠的耐缺氧能力。Objective:To optimize enrichment and purification technology of total flavonoids from Jinhe Yangxin Decoction and study the effect of it on the ability of anoxia endurance in mice. Methods:With rutin as reference substance,the content of total flavonoids was determined by aluminium nitrate chromogenic method,optimum macroporous resin by static adsorption/desorption and dynamic adsorption/desorption,and single factor tests were adopted to investigate effects of sample amount,sample flow rate,elution rate and other factors on purification technology;And observed the effect of total flavonoids from Jinhe Yangxin Decoction on the ability of anoxia endurance in mice by using the anoxia endurance on normal pressure test. Results:Compared with other resins,AB-8 macroporous resin showed the best separation efficiency,and the optimum purification technology was as follows:the concentration of sample solution of 0.5 g·m L-1,sampling velocity of 2 BV·h-1,sample volume of 2 BV,adsorption time of 4 h,eluted by 1.5 BV deionized water and 3 BV 50% ethanol at the speed of 2 BV·h-1,eluate was collected. Under these process conditions,the content of total flavonoids increased from 25.01% to 85.44%;And compared with the blank group,total flavonoids from Jinhe Yangxin Decoction could obviously prolong the living time of oxygen deficit-tolerance. Conclusion:AB-8 macroporous resin had a good purification effect of total flavonoids from Jinhe Yangxin Decoction,and it could obviously improve the ability of anoxia endurance.
