摘 要:目的探讨金不换总生物碱(JB-2)对胃粘膜的保护、抗炎及镇痛作用。方法将60只KM小鼠分为正常组(给予生理盐水)、模型组(给予生理盐水)、JB-2低、中、高三个剂量组(25、50、100 mg/kg)和三九胃泰组,连续灌胃给药3 d后,用乙醇(0.1 mL/10 g)进行灌胃,观察JB-2对模型小鼠溃疡指数及充血程度的影响。将60只SD大鼠分为正常组(给予等体积生理盐水)、模型组(给予等体积生理盐水)、JB-2低、中、高三个剂量组(15、30、60 mg/kg)和三九胃泰组,预给药7 d后,同样用乙醇(0.2 mL/100 g)灌胃进行造模,观察JB-2对模型大鼠溃疡指数及充血程度的影响,并检测相关氧化指标超氧化物歧化酶(SOD)和丙二醛(MDA)。分别取50只KM小鼠,随机分为模型组(给予生理盐水)、金不换总生物碱(JB-2)低、中、高三个剂量组(25、50、100 mg/kg)和阿司匹林组,每天灌胃给药1次,连续给药3 d后,分别以二甲苯致炎实验检测小鼠两耳片重差和醋酸致痛法记录小鼠15 min内的扭体次数,分别考察JB-2的抗炎、镇痛作用。结果在乙醇致大/小鼠急性胃粘膜损伤实验中,与模型组相比,JB-2中、高剂量组大/小鼠溃疡指数及出/充血程度明显减缓,大鼠SOD含量升高,MDA含量下降,阳性药对各指标也有明显改善,差异均有统计学意义(P<0.05)。与模型组比较,JB-2各给药组小鼠两耳片重差以及扭体次数明显减少,阳性药该指标也有明显改善,差异均有统计学意义(P<0.05)。结论金不换生物碱具有良好的胃粘膜保护、抗炎以及镇痛作用。Objective To investigate gastric mucosa-protective, anti-inflammatory, analgesic effects of total alkaloid of Stephania delavayi Diels (JB-2) in rats/mice. Methods 60 KM mice were randomly divided into control group, model group, JB-2 low- dose, medium- dose and high- dose groups (25, 50, 100 mg/kg, respectively) and Sanjiuweitai group. After three consecutive days of intragastric administration, the mice were gavaged with ethanol (0.1 mL/10 g), and the index of gastric mucosal lesion and and the degree of hyperemia in the model mice were observed. 60 SD rats were randomly divided into control group, model group, JB-2 low-dose, medium-dose and high-dose groups (15,30, 60 mg/kg, respedively)and Sanjiuweitai group. After pre-administration for 7 days, rats were also gavaged with ethanol (0.2 mL/100 g). The index of gastric mucosal lesion and and the degree of hyperemia were observed, and the content of SOD and MDA were detected. 50 KM mice were randomly divided into model group, JB-2 low-dose, medium-dose and high-dose groups (25,50,100 mg/kg, respectively) and aspirin group. They were given relevant medicine intragastrically, once a day, for consecutive 3 d. The xylene-induced inflammation test was used to detect the weight-loss of two ears of mice, and the acetic acid induced pain method was used to determine the number of writhing within 15 rain ; anti-inflammatory and analgesic effects of JB-2 were investigated. Results Compared with the model group, the index of gastric mucosal lesion and and the degree of hyperemia in rats/mice of JB-2 medium and high dose groups were significantly decreased. And the contents of SOD in JB-2 medium and high dose groups were significantly increased while MDA were significantly decreased. There was a significant improvement in the index of gastric mucosal lesion and the degree of hyperemia in Sanjiuweita group.
