摘 要:FT(FLOWERING LOCUS T)基因是植物多个开花途径的整合因子,在花发育中起重要作用。以水曲柳雌雄花发育转录组中FT序列为基础,通过PCR扩增获得该基因全长,命名为FmFT。其开放阅读框(ORF)为525 bp,编码174个氨基酸。生物信息学分析结果表明,其相对分子质量为19 795.2,等电点为5.90,是亲水性不稳定酸性蛋白;无跨膜区域,全部位于膜外;有1个PEBP保守结构域。同源比对结果显示,水曲柳FT基因编码的氨基酸序列与20个物种的FT蛋白序列同源性在83%~88%之间,与苹果(Malus domestica)同源性最高,为88%。系统进化结果显示,水曲柳FT蛋白单独聚为一类,与其他物种亲缘关系均较远。FmFT在水曲柳不同发育时期的雌雄花中的表达结果表明,在减数分裂时期和成熟孢子时期,雌雄花之间表达量差异显著。在雄花中,在减数分裂时期表达量达到最大值;在雌花中,表达量持续升高,在成熟胚囊时期达到最高值,是孢原细胞-大孢子母细胞时期的35.97倍,表明FmFT不仅在水曲柳雌雄花的发育中起着重要作用,且功能不同。FT (FLOWERING LOCUS T) gene is an integration factor in multiple flowering pathways, which plays an important role in flower development. Based on the FT sequence in the male and female transcriptome of Fraxinus mandshurica, the full length of the gene was obtained by PCR amplification and named as FmFT. Its open reading frame (ORF) was 525 bp, which encoded 174 amino acids. Bioinformatics analysis of FmFT showed that the relative molecular mass was 19 795.2 with the isoelectric point of 5.90, it was a hydrophilic and unstable acidic protein, and there was no transmembrane region and all were located outside the membrane. Besides, FmFT had a PEBP conserved domain. The results of homologous alignments showed that the homology of the amino acid sequences of Fraxinus mandshurica FT gene and the FT protein sequences of 20 species was between 83% and 88%, and the homology of the amino acid sequences of Fraxinus mandshttrica FT gene and the amino acid sequence encoded by Malus domestiea FT gene was the highest, which was 88%. Phylogenetic evolution showed that the FT protein of Fraxinus mandshurica was clustered into one kind, and the phylogenetic relationship was far from that of other species. Finally, the expression of FmFT in male and female flowers at different developmental stages of Fraxinus mandshurica showed that there was significant difference between male and female flowers during the meiosis and mature spores. The expression of the meiosis stage was the highest in male flower development, and the expression continued to rise until the mature embryo stage in female flower development, which was 35.97 times of sporozoite-megaspore mother cell, indicating that FmFT played important and different role in the development of male and female flowers in Fraxinus mandshurica.
