摘  要:为了建立金莲花的组织培养技术体系,为金莲花野生资源的保护和开发利用提供理论和技术支持,本试验以金莲花无菌播种苗为基础试验材料,以MS(1/2 MS)为基本培养基,在此基础上添加不同浓度的6-BA、NAA、IAA以及抗氧化剂,筛选适合金莲花组织培养各个阶段的最适培养基配方。结果表明,以金莲花无菌播种苗为初始材料,先经过壮苗培养是最佳的离体培养过程,壮苗培养的培养基配方是MS+6-BA4.0 mg/L+NAA 0.2 mg/L,丛生芽增殖的最佳培养基配方为MS+6-BA 4.0 mg/L+2,4-D 0.2 mg/L,抑制褐化的培养基配方为MS+6-BA 4.0 mg/L+2,4-D 0.2 mg/L+Vc 0.03 g/L,试管苗生根的最佳培养基配方为1/2 MS+IAA 0.5 mg/L,移栽炼苗的最好基质是草炭土,其移栽成活率达到了94.2%。The aim of this study was to establish the tissue culture system and provide the theoretical and technical support for the protection and exploitation of the wild resource of TroUius chinensis. Based on the aseptic seedling ofTrollius chinensis, the experiment took MS (1/2 MS) as basal medium and added in different concentrations of 6-BA, NAA, IAA and antioxidants, to screen the optimum formula for each stage of tissue culture. The result showed that taking the aseptic seedling as the initial material, after in vitro culture seedling is the best. The optimum culture medium of strong seedling was MS+6-BA 4.0 mg/L+NAA 0.2 mg/L, and the optimum shoot multiplication medium was MS+6-BA 4.0 mg/L+2,4-D 0.2 mg/L. Then the best browing inhibition medium was MS+6-BA 4.0 mg/L+2,4-D 0.2 mg/L+Vc 0.03 g/L, and the optimum medium for rooting was 1/2 MS+IAA 0.5 mg/L. Moreover, the best base material for transplanting seedling was peat in which the transplanting survival rate reached 94.2%.
