摘 要:本研究分析了在加有不同浓度H_2O_2的B5培养基上生长的杜仲愈伤组织总黄酮含量变化及过氧化物酶(POD)活性的变化及其两者的相关性。结果表明,5~10 mmol/L H_2O_2培养杜仲愈伤组织10 d时总黄酮含量最高。当杜仲愈伤组织培养5~10 d、外源H_2O_2浓度为10 mmol/L时的POD活性最高。相关性分析表明,杜仲愈伤组织总黄酮含量与POD活性之间的相关系数为r=0.420 61,具有一定的相关性,推测5~10 mmol/L H_2O_2可通过提高杜仲愈伤组织中POD酶活性,刺激黄酮的积累。本研究为杜仲黄酮的代谢调控及机制研究及在杜仲愈伤组织中获取高含量类黄酮的杜仲突变体提供科学依据。This study analyzed the changes of the flavonoid content and the peroxidase(POD) activity in the callus of Eucommia ulmoides cultured in B5 medium with different concentrations of H2O2 and their correlation. The results showed that the flavonoid content in the callus of Eucommia ulmoides was the highest with 5-10 mmol/L H2O2 for 10 d culture. The POD activity was the strongest when the callus were cultured for 5-10 d with 10 mmol/L H2O2. The correlation analysis indicated that the correlation coefficient was r =0.420 61 between the flavonoid content in callus and the POD activity, which implied the flavonoid content and the POD activity had a certain correlation. It was speculated that 5-10 mmol/L H2O2 stimulated the accumulation of flavonoids by improving the POD activity of the calli. This study provided the scientific basis for the study of metabolic regulation and mechanism of mutants of Eucommia ulmoides flavonoids and obtain high content of flavonoids in the callus.
