摘 要:海南岛益智及其姜科资源丰富,本研究基于ITS序列分析技术来研究海南岛不同居群益智及其姜科资源的遗传多样性。在海南岛内采集23份样品,其中益智样品18份,其他姜科植物样品5份,并从GenBank中获取3份山姜属植物的ITS序列作为研究系统的外群。通过软件Mega5.1比较分析了益智及其姜科植物的遗传距离和ITS序列的变异位点和信息位点,构建了NJ系统树等分析了益智种内与其姜科植物种群间的遗传多样性。研究结果表明海南岛不同居群的益智种内ITS序列无差异,未发现遗传变异位点,而姜科植物种间差异显著,表明其遗传多样性丰富。本研究为南药益智的种质资源的保护和可持续利用奠定理论基础。The res ources of Alpinia oxyphylla and other Zingiberaceae in Hainan Island are very rich, and this study is based on ITS sequence analysis techniques to study the genetic diversity of Alpinia oxyphylla and other Zingiberaceae in different geographical populations. 23 samples were collected from Hainan island, containing 18 samples of Alpinia oxyphylla samples and 5 samples of other Zingiberaceae plants. The ITS sequences of three Alpinia were obtained from Gen Bank as the outcrops of the research system. The software Mega5.1 was used to compare the genetic distance of the Alpinia oxyphylla and other Zingiberaceae plants and the mutation locus and information locus of the ITS sequence, and the NJ system tree was constructed to analyze the genetic diversity among the species of the Alpinia oxyphylla and among Zingiberaceae plant population. The results showed that there were no differences in the ITS sequences of Alpinia oxyphylla in the different geographic populations of Hainan island, and no genetic variation sites were found, but there were significant differences among the population of Zingiberaceae plant which meant abundant genetic diversity. This study might lay the theoretical foundation for the protection and sustainable utilization of southern medicine Alpinia oxyphylla germplasm resources.
