左归丸含药血清对大鼠卵巢颗粒细胞Smad4及周期蛋白Cyclin A表达的影响

摘　　要：目的:探讨Smad4蛋白对大鼠卵巢颗粒细胞细胞周期蛋白cyclin A的影响以及左归丸的干预作用。方法:以不同剂量的左归丸含药血清作用于体外培养的大鼠卵巢颗粒细胞,用MTS法检测各组颗粒细胞24、48、72 h增殖情况以及用Western bolt法检测卵巢颗粒细胞Smad4和cyclin A蛋白表达。结果:左归丸含药血清对大鼠卵巢颗粒细胞的增殖有促进作用,在作用48 h,2.5%左归丸含药血清组作用最为显著(P<0.05);同时Smad4和cyclin A蛋白也达到最大表达(P<0.05)。结论:左归丸含药血清对卵巢颗粒细胞增殖的促进作用与增加Smad4信号通路细胞周期蛋白cyclin A表达,抑制细胞分裂有关。Objective ： To study the effect of Smad4 protein on cycle proteins cyclin A and intervention func- tion of serum containing Zuogui Wan. Methods： Different doses of serum containing Zuogui Wan were used to culture rats＇ ovarian granulosa cells in vitro, the proliferation of ovarian granulosa cells was detected by MTS method after culturing 24,48, 72 h and Western blot was used to determine Smad4 and cyclin A protein expres- sions. Results： Serum containing Zuogui Wan could promote the proliferation of rats＇ ovarian granulosa cells in vitro ,the effect of 2.5% serum containing Zuogui Wan at 48 h was the best; meanwhile the expressions of Smad4 and cyclin A protein got the maximum （ P 〈 0. 05）. Conclusion ： Serum containing Zuogui Wan can promote the proliferation of rats＇ ovarian granulosa cells , that in related to raising the expressions of Smad4 and cyclin A pro- tein, inhibiting division of cells.
