莲慈颗粒对5/6肾切除大鼠肾小球硬化的抑制作用

摘　　要：目的探讨莲慈颗粒对5/6肾切除大鼠肾小球硬化的作用及其机制。方法将40只大鼠随机均分为假手术组(A组)、模型组(B组)、盐酸贝那普利治疗组(C组)和莲慈颗粒治疗组(D组)。光镜下观察肾脏病理变化,检测24-h尿蛋白定量、BUN、SCr水平和转化生长因子β1(TGF-β1)表达。结果与B组相比,C组和D组24-h尿蛋白定量、BUN、SCr水平和TGF-β1系膜区表达降低,肾脏病变改善(P<0.05或P<0.01);而C组与D组间各指标比较差异均无统计学意义(P>0.05)。结论莲慈颗粒可显著减轻肾纤维化,降低5/6肾切除大鼠肾组织TGF-β1表达,延缓肾小球硬化的进展。Objective To investigate the effect and underlying mechanism of Lianci granule on glomerulosclerosis in the rats with 5/6 nephrectomy.Methods Forty rats were equally randomized into four groups of A（sham operated）,B（5/6 nephrectomy model）,C（treated with benazepril hydrochloride 6 mg·kg^（-1）·d^（-1））and D（treated with Lianci granule 3 g/2 ml）.The pathological changes of the kidney were observed under light microscope.The quantity of 24-h urine protein,levels of BUN and SCr and expression of TGF-β1 were detected.Results Compared with group B,the quantity of 24-h urine protein,levels of BUN and SCr and expression of TGF-β1 were decreased and renal lesions were obviously improved in groups of C and D（P〈0.05 or P〈0.01）.There was no statistic difference in all indicators between groups of C and D（P〉0.05）.Conclusion Lianci granule can significantly reduce renal fibrosis,downregulate TGF-β1 expression and delay glomerulosclerosis development in the rats with 5/6 nephrectomy.
