摘 要:目的探讨乌司他丁对脂多糖诱导的大鼠急性肺损伤组织中水通道蛋白1(AQP-1)和Fas表达的影响。方法将60只大鼠随机均分为脂多糖组(A组)、乌司他丁组(B组)、脂多糖+乌司他丁组(C组)和对照组(D组),采用免疫组织化学染色检测各组肺组织中AQP-1和Fas的表达。结果 B组、C组和D组AQP-1表达均高于A组(0.542±0.035、0.499±0.039和0.593±0.084vs.0.218±0.072)(P<0.01),而Fas表达均低于A组(0.088±0.043、0.081±0.029和0.098±0.062vs.0.412±0.032)(P<0.01)。B组、C组和D组间AQP-1和Fas表达差异均无统计学意义(P>0.05)。结论乌司他丁可能通过上调AQP-1表达和下调Fas表达来减轻脂多糖诱导的大鼠急性肺损伤。Objective To investigate the effect of ulinastatin(UTI)on the expressions of aquaporin-1(AQP-1)and Fas in lung tissues of rats with acute lung injury(ALI)induced by lipopolysaccharide(LPS).Methods Sixty rats were equally randomized into four groups of A(intraperitoneal injection of LPS 3 mg/kg),B(intraperitoneal injection of UTI 10×10~4 U/kg),C(intraperitoneal injection of UTI 10×10~4 U/kg followed by LPS 3 mg/kg)and D(intraperitoneal injection of normal saline).The expressions of AQP-1 and Fas in lung tissues were detected by immunohistochemical staining.Results The expression of AQP-1 in groups of B,C and D was higher than that in group A(0.542±0.035,0.499±0.039 and 0.593±0.084 vs.0.218±0.072)(P〈0.01),while which of Fas was lower than that in group A(0.088±0.043,0.081±0.029 and 0.098±0.062 vs.0.412±0.032)(P〈0.01).There were no significant differences in the expressions of AQP-1 and Fas among groups of B,C and D(P〈0.05).Conclusion UTI can attenuate ALI induced by LPS in rats probably via upregulating AQP-1 expression and downregulating Fas expression.
