上转发光免疫层析技术检测梅毒心磷脂抗体的研制及初步评价

摘　　要：目的研制检测梅毒心磷脂抗体的上转发光免疫层析技术（UPT-LF）,并对其检测性能进行初步评价。方法制备心磷脂抗原,使其可以与UPT-LF结合。采用双抗原夹心原理,用上转发光纳米颗粒（UCP-NPs）标记的免疫层析技术进行检测。通过检测98份患者血清,比较UPT-LF、酶联免疫吸附试验（ELISA）和甲苯胺红不加热血清试验（TRUST）三种梅毒心磷脂抗体的检测方法并进行初步评价。结果氧化后的心磷脂保留了抗原性,且可与UPT-LF结合。建立了梅毒心磷脂抗体的上转发光免疫层析技术。三种检测心磷脂抗体的方法中,UPT法与ELISA法的真阳性率是66.7%,真阴性率是71.7%,CORREL值是0.702;UPT法与TRUST的真阳性率是80.6%,真阴性率是74.2%,CORREL值是0.577。结论上转发光免疫层析技术是检测梅毒心磷脂抗体的新方法,与基础实验ELISA法和TRUST相关性较好。Objective To develop the up-converting phosphor technology based on lateral flow assay（UPT-LF）to detect the phospholipid antibody of syphilis,and evaluate its detection performance preliminarily.Methods The cardiolipin antigen was prepared to combine with UPT-LF.The double antigen sandwich method and the immunochromatograghy were used with up-converting phosphor nana-particles（UCP-NPs）as the biological tracer to detect the syphilis cardiolipin antibody.98 serum samples of syphilis antibodies were detected by UPT-LF,and its results were compared with the same samples detected by enzyme linked immunosorbent assay（ELISA）and toluidine red unheated serum test（TRUST）.Results After oxidation,the cardiolipin retained the antigenicity and could be combined with UPT-LF.Then the UPT-LF for the phospholipid antibody of syphilis was established.The true positive rate,true negative rate and CORREL of UPT and ELISA was 66.7%,71.7% and 0.702 respectively.The true positive rate,true negative rate and CORREL of UPT and TRUST was 80.6%,74.2% and 0.577 respectively.Conclusion The UPT-LF is a new method to detect the phospholipid antibody of syphilitic,which is associated with the basic experimental ELISA and TRUST.
