摘 要:目的:观察苦参碱对人结肠癌SW480和SW480/M5细胞增殖、克隆形成、迁移侵袭及AKT/GSK3β/β-catenin信号通路的影响。方法:采用MTT法检测细胞增殖活性;克隆形成实验检测细胞克隆形成能力;Transwell实验检测细胞的迁移和侵袭能力;Western blotting检测AKT、GSK3β和β-catenin及各自磷酸化蛋白水平。结果:苦参碱呈浓度及时间依赖性抑制SW480和SW480/M5细胞的增殖,在24、48、72 h对SW480细胞的半数抑制浓度依次为1 919、832.8、318.1μg/mL,对SW480/M5细胞的半数抑制浓度依次为2 417、947、581μg/mL。苦参碱呈浓度依赖性降低SW480和SW480/M5细胞的克隆形成率,苦参碱可抑制SW480和SW480/M5细胞的迁移和侵袭能力,800μg/mL苦参碱能抑制SW480和SW480/M5细胞内AKT和GSK3β蛋白磷酸化,诱导β-catenin的磷酸化降解。结论:苦参碱可抑制结肠癌SW480和SW480/M5细胞的增殖、克隆形成和迁移侵袭能力,其机制可能与抑制AKT/GSK3β/β-catenin信号通路有关。Objective: To investigate the effect of matrine on proliferation,colony formation,migration,invasion and AKT/GSK3β/β-catenin signaling pathway of SW480 colon cancer cells. Methods: The cell proliferation was detected by MTT assay. The cell colony formation ability was assessed by colony formation assay. The ability of migration and invasion was evaluated by Transwell assay. The protein expressions of AKT,GSK3β,β-catenin and phosphorylation of protein were analyzed by Western blotting. Results: Matrine could inhibit the proliferation of SW480 cells and SW480/M5 cells in a concentration and time-dependent manner. The IC50 of SW480 cells were 1 919,832. 8 and 318. 1 μg/mL at 24,48 and 72 h,and the IC50 of SW480/M5 cells were 2 417,947 and 581 μg/mL. The colony formation of SW480 and SW480/M5 cells was decreased by matrine in a concentration-dependent manner,and matrine can inhibit the migration and invasion of SW480 cells and SW480/M5 cells. 800 μg/mL matrine inhibited the phosphorylation of AKT and GSK3β protein of SW480 and SW480/M5 cells,and induced the phosphorylation degradation of β-catenin. Conclusion: Matrine can inhibit the proliferation,colony formation,migration and invasion of SW480 cells and SW480/M5 cells,and the mechanism may be related to the inhibition of AKT/GSK3β/β-catenin signaling pathway.
