摘 要:目的:研究鸡血藤水提物的抗抑郁作用,并对其相关机理进行分析。方法:采用经典的小鼠悬尾实验和强迫游泳实验,观察鸡血藤水提物的抗抑郁作用。通过去势结合慢性不可预知性温和刺激复制雌性大鼠抑郁症模型,并从神经炎症的角度探讨鸡血藤水提物的抗抑郁作用机理。结果:鸡血藤水提物生药剂量8、4、2 g/kg均能显著缩短小鼠悬尾不动时间和游泳不动时间(P<0.05或P<0.01),而对小鼠自主活动无明显影响(P>0.05)。在抑郁大鼠实验中,鸡血藤水提物4 g/kg能显著缩短抑郁大鼠游泳不动时间(P<0.05)。鸡血藤水提物4、1 g/kg均能显著降低抑郁大鼠海马组织TNF-α和IL-1β水平(P<0.01),而对皮层组织TNF-α和IL-1β水平无明显影响(P>0.05)。免疫组化结果显示鸡血藤水提物4、1 g/kg能减少抑郁大鼠皮层和海马中NF-κBp65的表达。结论:鸡血藤水提物具有显著的抗抑郁作用,其作用机理可能与神经炎症的抑制有关。Objective: To investigate the antidepressive-like effect of aqueous extract from Spatholobus suberectus,and to study its related mechanism. Methods: The classical mice tail suspension test and mice force swimming test were used to evaluate the antidepressive-like effect of aqueous extract from Spatholobus suberectus. Female rat climacteric depression model was copied using the method of ovariectomy combined with chronic unpredictable mild stimulation( CUMS). The antidepressive-like mechanism of aqueous extract from Spatholobus suberectus was investigated from the view of nerve inflammation. Results: Aqueous extract from Spatholobus suberectus significantly decreased immobility time in tail suspension test and force swimming test in the doses of 8,4,and 2 g/kg( P〈0. 05 or P〈0. 01).However,it had no effects on autonomic activities. In depression rats experiment,aqueous extract of Spatholobus suberectus in the dose of4 g/kg significant decreased immobility time of depression rats in force swimming test( P〈0. 05). Aqueous extract from Spatholobus suberectus in the dose of 4,1 g/kg can significantly decreased the levels of TNF-α and IL-1β in hippocampus tissue of depression rats( P〈0. 01),while it has no obvious effects in cerebral cortex( P〈0. 05). Immunohistochemical results showed that the expression of NF-κBp65 in cerebral cortex and hippocampus tissue of depression rats were significantly decreased in the dose of 4,1 g/kg of aqueous extract of Spatholobus suberectus. Conclusion: Aqueous extract from Spatholobus suberectus has definitely anti depressive effect. The mechanism may be related to the inhibition of nerve inflammation.
