高效液相色谱法测定人血浆中舒乐安定的浓度

摘　　要：本文应用高效液相色谱法测定人血浆中舒乐安定的浓度。检测波长为240nm,利眠宁为内标,甲醇:磷酸盐缓冲液为移动相。回收率为97.3～108.9%,日内和日间误差(浓度0.60μg/ml)(CV)分别为2.5%和2.9%(n=9)。对临床数十名患者血浓测定,其血浓在0.4～0.87μg/ml,为临床合理用药提供了依据。Estazolam concentration in human plasma was determined by reverve phase HPLCwith UV detector at 240 nm.Chlordiazepoxide was used for internal standard. Therecorvery was 97.3～108.9%, the within day and day-to-day CV was 2.5% and 2.9%(n=9) at concentration range of 0. 15～0.60μg/ml, respectively. The method was sensitive to 0.05μg/ml plasma. The estazolam levels in 10 pateints were analyzedby this method. The plasma drug levels were at 0.40～0.87μg/ml. The result prov-ided a scientific basis for Clinical therapy.
