摘 要:目的 探讨黄芪总苷对染镉大鼠免疫功能及酶活性的影响。方法 将36只大鼠随机均分为空白组、模型组、黄芪总苷组。末次给药24 h后处死大鼠,计算胸脾指数。MTT法测定脾淋巴细胞的转化功能和NK细胞的杀伤活性,比色分析法测定大鼠脾脏中SOD、MDA的活性及大鼠血清中AST、ALT、LDH的活性,用ELISA法测定大鼠脾脏组织中IL-2、TGF-β1的含量。结果 与空白组比较,模型组的体重、胸脾指数、SOD的活性、IL-2的含量、淋巴细胞转化能力和NK细胞杀伤能力均明显降低;但模型组MDA、TGF-β1的含量及AST、ALT、LDH的活性均明显升高。与模型组比较,黄芪总苷组的体重、脾脏指数、SOD的活性、IL-2的含量、淋巴细胞转化能力和NK细胞杀伤能力明显升高,而MDA、TGF-β1的含量及AST、ALT、LDH的活性均明显降低。结论 黄芪总苷对镉所致大鼠的酶活性和免疫损伤具保护作用。OBJECTIVE To study the effects of astragalosides on the immune function and enzyme activity in rats induced by cadmi- um. METHODS 36 SPF Wistar rats were randomly divided into three groups : Blank group, cadmium - exposed group, and astragalo- sides group. After rats were sacrified, the indexes of thymus and spleen were calculated by weighing method. The transformation ability of T lymphocyte and the killing activity of NK cells were measured by MTT. The levels of SOD, AST, ALT, LDH and MDA were detected by colorimetrie assay. The contents of IL -2 and TGF -β1 were analyzed by ELISA. RESULTS Compared with the blank group, the weight and the indexes of thymus and spleen were decreased significantly in the cadmium - exposed group, the SOD activity and the IL- 2 content were also decreased significantly in the group. Meantime, the transformation ability of T lymphocyte and the killing activity of NK cells were reduced obviously in the cadmium - exposed group, while the levels of MDA,TGF - 131 , AST, ALT, LDH were increased. Compared with the cadmium - exposed group, the weights and the spleen index were both risen significantly in astragalosides treated group and the SOD activity and the IL -2 content were also improved significantly. The transformation ability of T lymphocyte and the killing activity of NK cells were enhanced obviously in astragalosides treated group. Yet the levels of MDA, TGF -β1, AST, ALT,LDH were lowered obviously in astragalosides treated group. CONCLUSION Astragalosides has protective effect on the immu- nity function damage and enzyme activity induced by cadmium in rats.
