摘　　要：目的 探究灵芝醇提取物在体内外的抗肿瘤活性。方法 用灵芝醇提取物处理6种人肿瘤细胞系（表皮癌A431、胃癌BGC823、直肠癌Caco2、卵巢癌ES-2、肺癌NCI-H460、结肠癌SW620）,观察细胞的生长形态、绘制生长曲线、进行MTT实验及平板集落形成实验。分别ig给予胃癌BGC823裸鼠50、100、150 mg·kg^（-1）灵芝乙醇提取物,计算瘤体积、抑瘤率及脏器系数。结果 体外实验中,灵芝提取物对表皮癌A431、胃癌BGC823、直肠癌Caco2、卵巢癌ES-2、肺癌NCI-H460、结肠癌SW620细胞的半数抑制浓度分别85.86、193.43、51.32、168.72、82.61、212.98μg·m L^（-1）。体内实验中,灵芝提取物50、100、150 mg·kg^（-1）的抑瘤率分别为32.6%、43.5%、47.2%。结论 灵芝提取物抑制表皮癌A431、直肠癌Caco2、肺癌NCI-H460细胞的增殖作用较强,且浓度大于40μg·mL^（-1）时,抑制增殖的作用较明显,体外还能抑制胃癌BGC823的生长。OBJECTIVE To explore the anti - tumor activities of the ethanol extracts of Garwderma lucidum （EEGL） in vitro and in vivo. METHODS Six human cancer cell lines including epidermoid cancer cells A431, stomach cancer cells BGC823, colorectal cancer cells Caco2, ovarian cancer cells ES - 2,lung cancer cells NCI - H460 and colon cancer ceils SW620 were treated with EEGL. The anti- tumor activities of EEGL was investigated with cellular morphology, growth curve, MTT assay and Colony - forming experiments. We established BGC823 stomach cancer nude mouse transplantation tumor model and treated with EEGL at the dose of 50, 100 and 150 mg.kg-l respectively,then the tumors＇ size,inhibitory rate and viscera coefficient were calculated. RESULTS In vitro experiment,the ICs0 of EEGL on epidermoid cancer cells A431, stomach cancer cells BGC823, colorectal cancer ceils Caco2,ovarian cancer cells ES -2,lung cancer cells NCI - H460 and colon cancer cells SW620 were 85.86,193.43,51.32,168.72,82.61,212.98 p^g.mL-lrespectively determined by MTT assay. In vivo experiment, the tumor inhibition rates of 50, 100,150 mg. kg-1 dose were 32. 6% ,43.5% and 47.2% ,respectively. CONCLUSION In vitro results imply that EEGL has inhibitory effect on epidermal cancer A431 ,rectal cancer Caco 2 and lung cancer NCI- H460. The inhibitory effect is obvious when the concentration was above 40 p^g＂ ml -l In vivo experiments indicate that EEGL can inhibit the growth of stomach cancer BGC823.
