摘 要:目的 建立何首乌与制何首乌的高效薄层色谱(HPTLC)指纹图谱,并对不同干燥方式、生长年限、产地来源的药材进行比较分析。方法 采用甲醇超声提取,浓缩滤液的方式提取样品。应用硅胶GF254高效预制薄层板,以三氯甲烷-甲醇-水(7∶3∶0.5)和三氯甲烷-丙酮-甲醇-水(6.5∶2.5∶1.5∶0.5)为展开剂,二次展开,置365 nm下检视。将所得薄层色谱图导入Chromap1.5色谱指纹图谱系统解决方案软件中生成共有模式,进行相似度分析。结果 何首乌与制何首乌的薄层色谱指纹图谱分别由7个与6个(不同产地制何首乌为5个)共有色谱峰组成。通过远红外干燥后的何首乌斑点信息丰富,整体较清晰;不同年限、产地的药材在成分上存在差异。结论 高效薄层色谱法可快速、简便、有效地鉴别何首乌与制何首乌,为评价药材内在质量的方法提供了依据。OBJECTIVE To establish a high performance thin layer chromatographic (HPTLC) fingerprints of Polygonum multiflo- rum Thunb. and its processed products, and to compare the fingerprints of products with the different drying methods, growing years and origins. METHODS The samples were extracted in methanol by ultrasonic and concentrated by methanol. The separation was performed on the pre -coated HPTLC GF254 silica gel plate. Second outspread system was used that chloroform -methanol -water(7 " 3 " 0.5) as the first system and chloroform - acetone - methanol - water(6.5 " 2.5 " 1.5 " 0.5 ) as the second, and the chromatograph was observed in 365 nm. The common patterns of HFFLC fingerprints were obtained through ( Chromapl. 5 ) solution software and the quality assessment were done by similarity analysis. RESULTS The common patterns of P. multiflorum and its processed products consisted of 7 characteristic peaks and 6 characteristic peaks ( processed products from different origins had 5 characteristic peaks). The ingredients number and content of P. multiflorum were changed by different drying methods, the samples drying by far - infrared were better than others. There were differences between samples with the different growing years and origins. CONCLUSION This method is rapid and convenient, and can effectively distinguish P. muhiflorum and its processed products, thus can be used for quality assess- ment of P. muhiflorum and its processed products.
