摘 要:目的:研究阿萨伊对非酒精性脂肪性肝炎(NASH)大鼠的保护作用及其健脾化浊功效。方法:设置空白组、模型组、水飞蓟宾组、葛根芩连汤组、阿萨伊高剂量组(2.4g/kg)、阿萨伊低剂量组(1.2g/kg)。空白组饲喂普通饲料,其余各组采用高脂饲料饲喂的方法制造NASH模型。分离血清,采用比色法检测血清总胆固醇(TC)、甘油三脂(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、白细胞介素(IL)-6、IL-10、肿瘤坏死因子-α(TNF-α)。肝组织匀浆,采用比色法检测肝脏过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、丙二醛(MDA),并使用HE染色法观察比较各组的肝脏病理情况,进行统计比较。结果:与模型组比较,阿萨伊高剂量组可以降低血清中ALT、AST、TC、TG、LDL-C、IL-6含量(P<0.05),升高血清中HDL-C含量(P<0.01);降低肝脏MDA含量(P<0.01),升高肝脏CAT含量(P<0.01),且对NASH大鼠的肝脏组织形态有改善作用。结论:阿萨伊对NASH大鼠具有一定的保护作用,实验结果可为探讨阿萨伊中药性能提供实验依据。Objective: To study the protective effect of Acai on non-alcoholic steatohepatitis (NASH) rats and its function of strengthening spleen and eliminating turbidity, Methods: The rats were divided into these six groups: the normal group, NASH model group, silybin group, Gegen Qinlian Decoction group, Acai high dose group (2.4g/kg), and Acai low dose group (1.2g/kg). Rats in the normal group were fed with common diet, and rats in other groups were fed with high-fat diet to establish NASH model rats, The serum was extracted, then the levels of these factors in serum were detected by colorimetry method, such as total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL-C), low density lipoprotein (LDL-C), alanine aminotransferase (ALT), aspartate aminotransferase (AST), interleukin-6 (IL-6), interleukin-10 (IL-10), and tumor necrosis factor-α (TNF-α ). The liver tissue homogenate was made to detect the levels of catalase (CAT), superoxide dismutase (SOD) and malondialdehyde (MDA) by eolorimetry method. The HE-staining was used to detect pathologic condition of liver in each group, and the statistical comparisons were done. Results: Compared with the model group, the contents of ALT, AST, TC, TG, LDL-C, IL-6 in serum of the Acai high dose group were decreased (P〈0.05), the content of HDL-C in serum was increased (P〈0.01), the content of MDA in liver was decreased (P〈0.01), the content of CAT in liver was increased (P〈0.01), and could significantly improve the morphology of liver tissue of NASH rats. Conclusion: Acai has some protective effect on NASH rats, and the experimental results can provide reference for discussing its properties of traditional Chinese medicine.
