微柱离心结合HPLC法测定鳖甲肽脂质体的包封率

摘　　要：目的:建立微柱离心结合高效液相(HPLC)测定鳖甲肽脂质体包封率的方法,筛选最佳的微柱离心条件,使脂质体与游离药物充分分离。方法:采用薄膜分散法制备鳖甲肽脂质体。采用葡聚糖凝胶Sephadex G-50微柱离心分离鳖甲肽脂质体和游离药物,HPLC法测定鳖甲肽浓度并计算包封率。结果:选择上样体积为0.6m L,然后1 000rcf离心3min,可以完全分离脂质体与外水相的药物;HPLC测得鳖甲肽脂质体平均包封率为32.23%(n=3)。结论:微柱离心可以达到将脂质体与游离药物很好的分离的目的,HPLC可以辅助测定脂质体中鳖甲肽的含量,这种方法可以用作鳖甲肽脂质体的分离并测定包封率的有效方法。Objective： To establish a method of minicolumn centrifugation combined with high performance liquid chromatography （HPLC） studies the encapsulation efficiency of Carapax Trionycis Peptide （HGRFG） in liposomes. Screening the best method of minicolumn centrifugation to separate the drug of HGRFG from the liposomes. Methods： HGRFG liposomes were prepared by thin-film dispersion method; then the liposomes and free drug were effective separated by the sephadex G-50 throw the method of rainicolumn centrifugation. The concentration of HGRFG in the liposome solution after demulsification by methanol could be accurately measured by the establish HPLC. Results： The optimum parameters of centrifugation method were that the volume of centrifugal was 0.6mL; the speed of centrifugation was 1 000rcf; the time of centrifugation was 3rain. The encapsulation efficiency of the drug of HGRFG liposome determined by this method was 32.23% （n=3）. Conclusion： The method of minicolumn centrifugation could achieve an effective separation between free drug and liposome and could apply to study the encapsulation efficiency of water-soluble drug in liposome. It could obtain the more exact results by the HPLC. The method is applicable for the determination of entrapment efficiency of HGRFG liposomes.
