摘  要:目的:探讨柴胡疏肝汤对外周血中促炎细胞因子诱导的色氨酸(TRY)代谢通路的影响。方法:设立空白组、模型组、氟西汀组和柴胡疏肝汤组,建立颞叶癫痫(TLE)相关性抑郁大鼠模型。在造模后第6周起,空白组和模型组不给予药物,氟西汀组和柴胡疏肝汤组分别给予氟西汀和柴胡疏肝汤治疗,观察治疗前和治疗过程中4周癫痫发作次数,外周血清IL-1β和IL-6表达,犬尿素/色氨酸(KYN/TRY)和五羟色胺/色氨酸(5-HT/TRY)比率,大鼠强迫游泳测试(FST)和糖水偏好测试(TPT)表现。结果:与模型组相比,柴胡疏肝汤组能够显著抑制外周血IL-1β和IL-6表达,降低KYN/TRY比率,减少强迫游泳测试中的不动时间和增加糖水偏好测试中糖精水摄取(P<0.05,P<0.01)。结论:柴胡疏肝汤通过抑制外周血IL-1β和IL-6表达,正常化TRY代谢通路,改善氟西汀抵抗的TLE相关性抑郁样行为。Objective: To discuss the effect of Chaihu Shugan Decoction (CHSGD) on periphery tryptophan (TRY) metabolic pathway induced by proinflammatory cytokine. Methods: Rats were randomly divided into naive group, control group, Fluoxetine (FLX) group and CHSGD group, and rat model of temporal lobe epilepsy (TLE) related depression was built. After sixth weeks, the naive group and control group were not given to drugs, rats in FLX group were received FLX treatment and rats in CHSGD group received CHSGD treatment. The seizure frequency were calculated in 4 weeks before and during treatment, the IL- l 13 and IL-6 expression, the kynurenine (KYN)/tryptophan(TRY) ratio and serotonin(5-HT)/TRY ratio, taste preference test (TPT) and forced swim test (FST) were evaluated. Results: In the CHSGD group, periphery IL-1 13 and IL-6 expression were inhibited notably, the KYN/TRY ratio decreased, the immobility time in FST reduced and the saccharin solution consumption in TPT increased when compared to control group (P〈0.05, P〈0.01). Conclusion: CHSGD improve temporal lobe epilepsy associated depressive-like behavior which is resisted to fluoxetine by inhibiting periphery IL-1 13 and IL-6 expression and normalizing periphery TRY metabolites.
