醒脑益智剂对2型糖尿病认知功能障碍小鼠性激素及ChAT、ApoE mRNA表达的影响

摘　　要：目的:研究醒脑益智剂对2型糖尿病(T2DM)认知功能障碍(CI)小鼠性激素和大脑海马及皮质中胆碱乙酰转移酶(Ch AT)、载脂蛋白E(Apo E)表达的影响,阐释"胰(脾)-脑-肾轴"消渴呆病病机学说。方法:10周龄雌性昆明小鼠为正常组,10周龄雌性KKAy糖尿病小鼠随机分为模型组、阳性组、中药组。阳性组予吡拉西坦片,中药组予醒脑益智剂,正常组和模型组予等体积0.9%氯化钠溶液。给药8周。给药前后各行1次Morris水迷宫。水迷宫结束后1周内测定性激素水平,ChAT和ApoE mRNA的表达。结果:与模型组比较,正常组、阳性组和中药组给药后第5天潜伏期明显减少(P<0.05),正常组、中药组LH、T下降(P<0.01,P<0.05),E2、P升高(P<0.01,P<0.05);与阳性组比较,中药组LH下降(P<0.05);与正常组比较,模型组海马内ChAT mRNA降低(P<0.05);与模型组比较,阳性组和中药组海马内Ch AT mRNA升高(P<0.05);与正常组比较,其余3组ApoE mRNA降低(P<0.05)。结论:糖尿病痴呆的出现可能与性激素通路有关,醒脑益智剂对糖尿病CI有明显改善作用,这一作用可能是通过提高E_2、P水平和大脑海马内ChAT mRNA表达水平,降低LH水平实现的。Objective： To study the effect of Xingnao Yizhi Prescription on the expressions of sex hormone, ChAT and ApoE of T2DM mice with cognition impairment, and imerpret the pathogenesis of dementia with consumptive thirst from pancreas （spleen）-brain-kidney axis. Methods： 10-week KM mice were used as normal group, while 10-week KKAy diabetic mice were randomly divided into model group, positive group and TCM group. Positive group was given piracetam, TCM group was given Xingnao Yizhi Prescription, normal and model groups were given equal volume of saline, the drug administration lasted for 8 weeks, Morris water maze test was measured before and after the experiment, and then the expressions of sex hormone, ChAT and ApoE were tested. Results： Compared with model group, the normal group, positive group and TCM group had shorter incubation period （P〈0.05）, Compared with model group, the normal and TCM group had lower LH and T levels （P〈0.01, P〈0.05）, and higher E2, P levels （P〈0.01, P〈0.05）. Compared with positive group, the TCM group had lower LH levels （P〈0.05）. Compared with normal group, the model group had lower ChAT mRNA level. Compared with model group, the positive group and TCM group had higher ChAT mRNA level （P〈0.05）. Compared with normal group, the KKAy groups had lower ApoE level （P〈0.05）. Conclusion： The emergence of diabetic dementia might be related to the sex hormone pathway. Xingnao Yizhi Prescription could improve the effect of mild cognitive impairment in patients with diabetes mellitus, because of its effects to improve plasma E2, P levels, and increase the level of estrogen receptor CHAT, decrease the level of LH in hippocampus.
