摘 要:目的:观察不同浓度淫羊藿苷含药血清对兔膝软骨细胞增殖率及糖胺聚糖(GAG)分泌的影响。方法:将3周龄健康新西兰大白兔后肢膝关节分离培养软骨细胞进行鉴定,用制备低、中、高浓度淫羊藿苷(ICA-L、ICA-M、ICA-H)及低剂量仙灵骨葆胶囊(XLGB)含药血清干预分离培养软骨细胞,形态学观察软骨细胞及MTT检测细胞增殖率,邻联甲苯胺(DMB)染色法检测细胞内GAG含量。结果:与对照组比较,空白血清组及含药血清组均使兔软骨细胞大量增殖(P<0.01);XLGB组和ICA-L组细胞增殖率较空白血清组显著增高(P<0.01),ICA-L组明显。与对照组比较,空白血清组及XLGB组、ICA-L组、ICA-M组均使兔软骨细胞GAG分泌增多(P<0.01),其中,ICA-L组分泌GAG最多。结论:低浓度淫羊藿苷含药血清对兔软骨细胞增殖率及其分泌GAG功能强大,单纯淫羊藿苷原液对软骨细胞生长有抑制作用,经过生物转化后的含药血清药效与浓度非正比关系。Objective: To observe the effects of icariin-containing serum on the proliferation and glycosaminoglycan (GAG) secretion in rabbit knee cartilage cell. Methods: Chondrocytes were isolated and cultured from hind limbs of healthy New Zealand white rabbits at 3 weeks of age. The chondrocytes were isolated and cultured with low, medium and high concentrations of icariin (ICA-L, ICA-M, ICA-H) and low-dose Xianling Gubao Capsule (XLGB) containing serum, respectively. The chondrocytes were observed morphologically and the proliferation of chondrocytes were detected by MTT. The intracellular GAG content was detected by DMB staining. Results: Compared with the control group, the ehondrocytes proliferated significantly in the blank group and the drug-containing serum group (P〈0.01). The proliferation rates of the XLGB group and ICA-L group were significantly higher, and the ICA-L group was the higher group. Blank group and XLGB group, ICA-L group and ICA-M group increased the GAG secretion of rabbit chondrocytes (P〈0.01), and the ICA-L group secreted the most GAG. Conclusion: ICA-L has the strongest function on the proliferation of rabbit chondrocytes and the secretion rate of GAG, followed by XLGB, and the ICA-H has inhibitory effect on cartilage cell secretion of GAG.
