摘 要:目的:观察平喘宣肺汤对支气管哮喘大鼠气道炎症反应的影响。方法:选取SPF级SD大鼠,分为正常组、模型组、地塞米松组、自拟平喘宣肺汤高、中、低剂量组,每组10只,采用卵清白蛋白致敏及雾化吸入激发的方法建立大鼠支气管哮喘模型。实验结束后对大鼠肺组织灌洗液(BALF)EOS、NEU、LYM、MAC计数,ELISA方法测定大鼠肺组织的IL-1、IL-6、NF-α水平,Western-bloting方法检测大鼠肺组织PKC、MMP-9蛋白表达。结果:自拟平喘肺汤可以降低EOS、NEU、LYM、MAC比率,与模型组比较有显著性差异(P〈0.05),自拟平喘宣肺汤可降低大鼠血清IL-1、IL-6、NF-α水平,与模型组比较有显著性差异(P〈0.05),可以减少肺组织PKC、MMP-9蛋白表达,且具有剂量依赖性。结论:自拟平喘宣肺汤可有效减轻支气管哮喘大鼠的炎症反应,可能通过调控PKC、MMP-9蛋白表达相关。Objective: To observe the effect of Pingchuan Xuanfei Decoction on airway inflammation in asthmatic rats.Methods: SPF SD rats were divided into normal group,model group,dexamethasone group,Pingchuan Xuanfei Decoction high,medium and low dose groups,10 rats in each group. We used ovalbumin sensitization and inhalation excitation method to establish rat model. After the end of the experiment in rat lung lavage fluid( BALF),EOS,NEU,LYM and MAC were counted and we determined lung tissues' IL-1,IL-6 and NF-α levels by ELISA method and the expressions of PKC and MMP-9 protein were measured by Western-bloting method. Results: Pingchuan Xuanfei Decoction can reduce EOS,NEU,LYM and MAC and there was significant difference compared with those of the model group( P 0. 05). Pingchuan Xuanfei Decoction can reduce serum IL-1,IL-6 and NF-α levels and there was significant difference compared with those of the model group( P 〈 0. 05) and can reduce the the expressions of PKC and MMP-9 protein in lung tissue in a dose-dependent manner. Conclusion: Pingchuan Xuanfei Decoction can effectively reduce the inflammatory response in asthmatic rats,which may be related to the regulation of PKC and MMP-9 protein expression.
