摘 要:为了研究繁殖候选基因ADAMTS-1对香猪繁殖性状的调控机理,本研究以高产、低产两个香猪群体为实验材料,克隆ADAMTS-1基因全长,分析其存在的多态性位点,并对第七外显子5996位进行产仔数的遗传效应分析,同时检测两个香猪群体中该基因的拷贝数变异。结果表明,ADAMTS-1基因包含8个内含子和9个外显子区域。该基因第七外显子5996位存在多态性,香猪品种中以B等位基因为主,基因型频率BB〉AB,未检测到纯合AA基因型的个体,而西方猪种中以AA或AB型为主。高产和低产两个香猪群体中的ADAMTS-1基因的拷贝数都是复制型为主,但存在拷贝数的差异,高产香猪群体中的拷贝数增加较多,多为3个或3个以上拷贝,而低产群体中多为双拷贝,低产群体中的拷贝数明显低于高产群体。结果提示,ADAMTS-1基因的拷贝数变异以及第7外显子5996位的多态性可能是影响香猪繁殖力的重要因素之一。In order to study the regulation mechanism of A DA MTS-1 for reproductive trait of flagrance pigs, the study cloned the full length of ADAMTS-1 in two groups of high yield and low yield fragrance pigs, analyzed its polymorphic loci and genetic effect of litter size of 5 996 site on the 7th exon in fragrance pigs. Furthermore, the study also identified the copy number variation between two fragrance pig colonies. The results showed that, the candidate gene ADAMTS-1 included 8 introns and 9 exons. 5 996 site on the 7th exon of ADAMTS-1 was polymorphic, and the dominant genotype in fragrance pig was BB whose genotype frequency was higher than that of AB and no homozygous AA was detected, while AA and AB were dominant in western pig breeds. Moreover, the copy numbers of ADAMTS-1 were almost replicative in 2 groups of flagrance pig population, but variation existed. Copy number of the large litter size population rised more of which most contained three or more copies, while two copies often occured in the small litter size population which were significantly lower than the high yield group. These results indicated that the copy number variation of ADAMTS-1 and its polymorphism in position 5 996 might play a critical role in determining the reproductive capacity of fragrance pig.
