摘  要:目的研究一种含5%(质量分数)纳米单质银颗粒的-磷酸三钙复合支架(β-TCP-Ag)的细胞毒性及在家兔体内的抗菌性和促成骨性能,从而为临床治疗骨髓炎合并大面积骨缺损提供新的思路。方法体外:以100%-磷酸三钙支架(β-TCP)作为对照组,在体外用Cell Counting Kit-8(CCK-8)试剂检测支架浸提液对大鼠骨髓间充质干细胞(r BMSCs)的细胞毒性;体内:选取新西兰大白兔28只,以β-TCP为对照,一期在4周内建立兔股骨骨髓炎模型,再二期植入支架后分别饲养4周和12周,然后取支架植入区域骨组织,用平板涂布法检测金黄色葡萄球菌菌落的相对个数;用显微电子计算机断层扫描(Micro-CT)观察支架植入区域骨组织长入情况。结果CCK-8检测显示各组间细胞相对活性未见明显差异,表明β-TCP-Ag在体外未见有明显细胞毒性;细菌平板涂布法显示植入β-TCP-Ag组材料周围骨组织培养金黄色葡萄球菌菌落数远小于β-TCP组;Micro-CT分析表明β-TCP-Ag组骨缺损区域骨量明显高于β-TCP组。结论含5%纳米银颗粒的-磷酸三钙复合支架具有良好的生物相容性和抗菌能力,在体内杀灭细菌后有利于骨缺损的愈合,是一种良好的抗菌和骨缺损植入物材料。在改进骨感染合并骨缺损的治疗中具有重要的潜能。Objective To investigate the antibacterial and osteogenic activity of 5% (mass fration) elemental silver nan- oparticle-incorporated p-tricalcium phosphate scaffold (p-TCP-Ag) as a new idea for treating osteomyelitis with large area of bone defect. Methods In vitro: the cytotoxicity of the p-TCP-Ag was characterized by Cell Counting Kit-8 assay in comparison to pure p-tricalcium phosphate scaffold. In vivo: 28 New Zealand rabbits underwent osteomyelitis of the left femoral condyle with staphylococcus aureus infection for 4 weeks, then were implanted scaffold (p-TCP-Ag or p- TCP). After 4 weeks' raising, the rabbits were sacrificed to collected the bone tissue around the scaffold, then the relativenumber of staphylococcus aureus was detected by spread plate method. After 12 weeks' raising, the rabbits were sacrificed and microcomputed tomography (Micro-CT) was used to observe the bone healing around the scaffold. Results The result of cytotoxicity test show that there is no significant difference of the relative cell number between the P-TCP-Ag group and the control group. The result of spread plate method show that the relative number of staphylococcus aureus in the p-TCP-Ag group is remarkable higher than that of the control group. The Micro-CT analysis show that the bone volume/t?tal volume (BV/TV) of p-TCP-Ag group is significant higher than that of the control group. Conclusion The above results indicates that this potential biomaterial for treating osteomyelitis with bone defect has good biocompatibility,antibacterial activity and osteogenic activity. This study demonstrates that the p-TCP-Ag with a combination of antibac-terial and osteogenic activity can be achieved for the reconstruction of bone defects while treating infections.
