摘  要:利用20 kHz超声波在功率分别为45、90和135 W时处理10、20、40、60和80 min辅助法提取牛皮胶原。结果表明超声波辅助处理能明显提高胶原提取率,但当超声功率达到135 W时会导致部分胶原发生降解,胶原的热变性温度随着超声功率的增强和超声时间的延长而有降低的趋势。优化得到牛皮胶原超声辅助提取的最佳超声条件为功率90 W、作用时间20 min,此时凝胶电泳、红外和紫外图谱均显示胶原三股螺旋结构未改变,胶原提取率为46.76%,比未超声处理的胶原提取率提高了13.58%。Collagen was extracted from bovine hide assisted by ultrasonic when the power of 20 kHz ultrasound respectively was 45, 90 or 135 W and the ultrasonic time was 10, 20, 40, 60 or 80 min.The results showed that the ultrasonic-assisted method could improve the extraction rate of collagen obviously. A amount of collagen began to degrading when the ultrasonic power was 135 W.The thermal denaturation temperature of collagen decreased slightly with the increase of ultrasonic power and ultrasonic time.In this experiment, the optimal ultrasonic conditions were 90 W and 60 to 80 min,at this point, the gel electrophoresis, infrared and ultraviolet spectra showed that the triplehelical structure of collagen was retained. And on this condition, the extraction rate of collagen was about 53.00%, compared with the extraction without ultrasonic assistance,increased by about 20.00%.
