摘  要:目的探讨miRNA-126和miRNA-155在系统性红斑狼疮(SLE)患者血浆中的表达及其临床意义。方法选取40例SLE患者为SLE组,并根据SLE疾病活动指数分为SLE活动期患者与SLE非活动期患者;另选择同期相近年龄段20例健康体检者作为对照组。采用实时荧光定量聚合酶链反应(qRT-PCR)技术检测所有患者血浆miRNA-126和miRNA-155表达水平并进行统计分析。比较对照组与SLE组、SLE活动期患者与SLE非活动期患者miRNA-126和miRNA-155表达水平。结果SLE组miRNA-126表达上调倍数为3.55±2.99,miRNA-155表达下调倍数为0.49±0.24,与对照组(1.03±0.25、1.01±0.16)比较差异均有统计学意义(P均〈0.05)。SLE组中SLE非活动期者miRNA-126上调倍数[2.57(2.03-3.10)]低于SLE活动期者[4.53(2.71-6.34)],差异有统计学意义(P〈0.05),SLE非活动期者miRNA-155下调倍数[0.47(0.22-0.63)]与SLE活动期者[0.44(0.18-0.74)]比较差异未见统计学意义(P〉0.05),表明miRNA-126与疾病活动程度成正相关,miRNA-155的表达与SLE疾病活动无关。miRNA-126和miRNA-155的ROC曲线下面积(AUC)分别为0.825(95%CI:0.697-0.953)和0.852(95%CI:0.744-0.859),在SLE患者中异常表达有较好的特异性及敏感性。结论 SLE患者血浆中miRNA-126和miRNA-155均表达异常,提示其参与了SLE的发病机制并在其中发挥重要作用,且两者可成为SLE辅助诊断标志物和有效治疗靶点。Objective To investigate the expression and clinical significance of miRNA-126 and miRNA-155 in systemic lupus erythematosus(SLE) patients.Methods Forty SLE patients were selected as SLE group and were divided into patients in SLE active stage or in SLE non-active stage; another 20 healthy people who are about same age as selected SLE patients were allocated to control group. The levels of plasma miRNA-126 and miRNA-155 were detected by real-time quantitative polymerase chain reaction (qRT-PCR) technique in all research objects, and the results were recorded and analyzed. The levels of miRNA-126 and miRNA-155 were compared both between SLE group and control group, and between patients in SLE active stage and in SLE non-active stage.Results There were significant differences in raising multiple of miRNA-126 and decreasing multiple of miRNA-155 between SLE group (3.55±2.99, 0.49±0.24) and control group (1.03±0.25, 1.01±0.16), all P〈0.05. The raising multiple of miRNA-126 of patients in SLE active stage was lower than that of patients in SLE non-active stage (P〈0.05); but there was no significant difference in decreasing multiple of miRNA-155 between patients in SLE active stage and in patients non-active stage (P〉0.05); the expression of plasma miRNA-126 was positively correlated with disease activity (P〈0.05), while the expression of plasma miRNA-155 was independent of SLE activity (P〉0.05). The area under the ROC curve of miRNA-126 and miRNA-155 was 0.825 (95%CI: 0.697-0.953) and 0.852 (95%CI: 0.744-0.859), respectively, that is, expression of miRNA-126 and miRNA-155 had good specificity and sensitivity in SLE patients.Conclusions The expression of miRNA-126 and miRNA-155 in the plasma of patients with SLE is abnormal, suggesting that mi-RNA-126 and miRNA-155 are involved in the pathogenesis of SLE and play important roles in the pathogenesis of SLE. Both of them can be used as auxiliary markers for diagnosis and effective therapeutic targets.
