补脾强力复方对EAMG大鼠血清CORT、E、ACTH、CRH干预作用的研究

摘  要:目的:观察补脾强力复方对实验性自身免疫性重症肌无力(EAMG)大鼠血清皮质醇(CORT)、肾上腺素(E)、促肾上腺皮质激素(ACTH)、促肾上腺皮质激素释放激素(CRH)的影响,探讨补脾强力复方治疗重症肌无力(MG)的可能机制。方法:采用鼠源性AchR-α亚基97-116肽段序列(R97-116)免疫接种雌性Lewis大鼠,建立EAMG大鼠动物模型;通过ELISA法检测EAMG大鼠免疫后第7周CORT、E、ACTH、CRH含量水平,比较补脾强力复方大、中、小剂量与强的松的干预效果。结果:与正常组比较,模型组大鼠血清ACTH、CRH水平显著升高(P<0.05),血清E水平显著降低(P<0.01),CORT水平差异无统计学意义(P>0.05);与模型组比较,补脾强力复方各剂量组及强的松组ACTH、CRH水平显著降低(P<0.01),血清E、CORT水平显著升高(P<0.01);补脾强力复方大剂量组血清ACTH、CRH、E水平更接近正常组水平。结论:补脾强力复方能调节EAMG大鼠血清CRH、ACTH、E、CORT的分泌,能有效降低EAMG大鼠血清ACTH、CRH水平及升高E、CORT水平,其大剂量组使各激素的分泌更接近正常组水平,这可能是补脾强力复方治疗MG的可能机制。Objective: To investigate the interventional effect of compound spleen-tonifying prescription on serum levels of corticosterone( CORT),epinephrine( E),adrenocorticotropic hormone( ACTH),and corticotropin-releasing hormone( CRH) in rats with experimental autoimmune myasthenia gravis( EAMG),as well as the possible mechanism of action of compound spleen-tonifying prescription in the treatment of myasthenia gravis( MG). Methods: The mouse-derived AchR-α subunit 97-116 peptide( R97-116) was used for immunization of female Lewis rats to establish a rat model of EAMG. ELISA was used to measure the levels of CORT,E,ACTH,and CRH at week 7 after immunization,and the interventional effect of high-,middle-,and low-dose compound spleen-tonifying prescription was compared with that of prednisone. Results: Compared with the normal group,the model group had significant increases in the serum levels of ACTH and CRH and a significant reduction in the serum level of E( P 0. 01),while there was no significant difference in the level of CORT( P 0. 05). Compared with the model group,the high-,middle-,and low-dose compound spleen-tonifying prescription groups and the prednisone group had significant reductions in the levels of ACTH and CRH and significant increases in the serum levels of E and CORT( P 0. 01); the high-dose compound spleen-tonifying prescription group had similar serum levels of ACTH,CRH,and E as the normal group. Conclusion: Compound spleen-tonifying prescription can regulate the secretion of CRH,ACTH,E,and CORT in serum,and effectively reduce the serum levels of ACTH and CRH and increase the levels of E and CORT,and high-dose compound spleen-tonifying prescription helps to achieve almost normal secretion of these hormones,which may be one of the mechanisms of action of compound spleen-tonifying prescription in the treatment of MG.
