摘 要:目的探讨外源性促红细胞生成素对压力超负荷引起小鼠左心功能障碍的保护作用。方法选取45只C57BL/6J雄性小鼠,主动脉弓缩窄术(TAC)后,将小鼠随机分为假手术组(Sham)、模型组(TAC-PBS)和治疗组(TAC-EPO),每组15只。采用M型超声心动图分别测量小鼠术前和术后2、4、6、8周左室舒张末期直径、左室收缩末期直径、左室短轴缩短率、舒张末期室间隔厚度、左室游离壁厚度和心室率;术后8周进行心导管测定,测量左心室收缩峰压、左室舒张末压和心室率。结果超声心动图示,与假手术组比较,模型组和治疗组小鼠左心室舒张和收缩末期直径均增加(P<0.05),左心室短轴缩短率均减少(P<0.05);但经EPO治疗后,与模型组相比,治疗组小鼠左心室舒张和收缩末期直径显著降低(P<0.05),左心室短轴缩短率显著增加(P<0.05)。结论外源性促红细胞生成素对压力超负荷引起的小鼠左心功能障碍的保护作用可能通过降低左心室扩张而实现。Objective To study if the exogenous erythropoietin protects left ventricle against pressure overloadinduced dysfunction in mice. Methods 45 C57BL/6J male mice were randomly divided into sham group(Sham), model group(TAC-PBS) and treatment group (TAC-EPO) after transverse aortic constrietion(TAC), 15 mice in each group. The left ventricular end-diastolic diameter, left ventricular end systolic diameter, left ventricular fractional shortening, end diastolic ventricular septal thickness, left ventricular free wall thickness and heart rate were measured by M type echocardiography. The cardiac catheterization was measured at 8 weeks after operation, and left ventricular peak systolic pressure measurement, Left ventricular end diastolic pressure and ventricular rate were detected. Results Compared with sham group, left ventrieular diastolic and systolic diameter of mice in model group and treatment group mice were increased(P〈0.05), left ventricular fractional shortening was decreased (P〈0.05). After EPO treatment, compared with model group, left ventricular diastolic and systolic diameter of mice in treatment group were decreased significantly (P〈0.05), left ventricular fractional shortening was increased significantly(P〈0.05). Conclusion The exogenous erythropoietin protects left ventricle against pressure overload-induced dysfunction in mice may be through ameliorating left veutricular dilatation.
