摘  要:目的探讨三七三醇皂苷(panax notoginseng,PTS)对糖尿病大鼠学习记忆的改善作用及可能机制。方法链脲佐菌素诱导SD大鼠糖尿病模型,将大鼠分为对照组、糖尿病组及PTS组,3个月后水迷宫实验检测大鼠学习记忆,Nissl染色观察脑海马区神经元形态,TUNEL染色观察海马区神经元凋亡,免疫组化染色和Western blot检测凋亡相关基因Bax在海马区的表达情况。结果与对照组相比,糖尿病组大鼠学习记忆能力减退,表现为逃避潜伏期长以及穿经水迷宫平台次数减少,而且脑海马区神经元形态不规则,排列紊乱,结构不清晰,凋亡神经元及Bax表达均明显增加。PTS组与对照组无明显差别。结论PTS改善糖尿病大鼠学习记忆能力可能与下调糖尿病脑海马区神经元Bax的表达,抑制神经元凋亡相关。Objective To explore the beneficial effect of panaxtrial saponins (PTS) on cognitive dysfunction of the diabetic SD rats. Methods SD rats were intraperitoneally injected with streptozotocin to induce diabetic models. Rats were divided into PTS, diabetic model and control groups. Learning and memory ability was observed by Water maze, TUNEL and Nissl staining were used to reveal the morphological and apoptosis alterations in hippocampal neurons, and the expression of apoptosis-related Bax in hippocampal was observed by immunohistochemistry and Western blot. Results Compared with control group, diabetic group showed cognitive dysfunction in water maze test, presenting increased escape latency and decreased cross number, together with increased apoptosis, disarranged neurons showing unclear structure, and up-regulation of Bax expression, while, PTS group showed no difference compared with control group. Conclusion Panaxtrial saponins ameliorating learning and memory ability might be related to the inhibition of Bax expression and apoptosis in hippocampal neurons of diabetic rats.
