GYY4137对大鼠肾缺血再灌注损伤后氧化应激及炎症反应的保护作用

摘  要:目的观察硫化氢缓释供体GYYdl37对大鼠肾脏缺血再灌注损伤(ischemicandreperfusioninjury,IRI)后氧化应激及炎症反应的作用。方法随机将32只SD大鼠分为假手术组、假手术+药物组、缺血组、药物组,每组8只,进行如下实验:①假手术组:切除大鼠右肾后缝合腹壁;②假手术+药物组:切除大鼠右肾后,腹腔注射GYY4137后缝合腹壁;③缺血组:切除右肾后,用无创伤血管夹夹闭左肾动、静脉45min进行缺血,然后开放血管夹进行再灌注;④药物组:手术步骤与缺血再灌注组相同,但在缺血后、再灌注之前,腹腔注射500μmol/LGYY413710mL。全自动生化分析仪检测大鼠血清肌酐和尿素氮,硫代巴比妥酸法和黄嘌呤氧化酶法分别测定血清脂质过氧化产物丙二醛(MDA)和超氧化物歧化酶(SOD),HE染色观察大鼠肾组织的病理改变,PCR检测TNF-α和IL-6的mRNA水平,蛋白免疫印迹法(WesternBlot)检测IL-2的含量。结果缺血组较假手术组和假手术+药物组相比,肾功能水平、MDA含量、病理损害程度、TNF-α、IL-6和IL-2表达量明显升高,而SOD水平明显降低;药物组与缺血组相比,肾功能水平、MDA含量、病理损害程度、TNF-α、IL-6和IL-2表达量明显降低,而SOD水平明显升高。结论GYY4137能减轻大鼠肾脏IRI损伤,其机制可能与抑制缺血再灌注损伤后的氧化应激及炎性反应有关。Objective To investigate the effect of GYY4137 on oxidative stress and inflam- mation caused renal ischemia and reperfusion injury ( IRI ) in rats. Methods A total of 32 SD rats were randomly divided into four groups, sham operation group, sham operation + drug group, IRI group and drug group. Each group had 8 rats. In sham operation group, the right kidneys of rats were ex- cised. In sham operation + drug group, the right kidneys of rats were excised and GYY4137 (500μ mol/L) was injected intraperitoneally. In IRI group, with no wound vascular clamp, left renal arteries and veins of rats were clamped for 45 min, then opened vascular clamp was opened and infused again. In drug group, operation steps were the same as the IRI group, but at the end of ischemia and the beginning of reperfusion, GYY4137 (500μmol/L) was injected intraperitoneally. The parameters ( blood urea nitrogen and creatine) of renal function were determined by auto - biochemiealanalyzer. The kidneys of rats were removed for determination of SOD activity and MDA concentration. The chan- ges of renal tissue were examined by a microscope. The expression of the TNF - α, IL - 6and IL - 2 were detected by PCR and Western Blot. Results Compared with sham operation group, BUN, Cr, MDA and the expression of TNF -α, IL- 6 and IL- 2 were significantly increased in IRI group. How- ever, these targets in drug group were significantly decreased compared with those in IRI group. TheSOD in IRI group was significantly decreased compared with those in sham operation group. Conclu- sions GYY4137 can alleviate oxidative stress and inflammation induced by renal IRI.
