摘 要:目的:探讨脑还丹对Aβ25-35诱导的痴呆模型大鼠炎性因子表达及海马区超微结构的影响。方法:原代海马神经元分为对照组、β-淀粉样蛋白(Aβ)组、脑还丹低剂量组、脑还丹高剂量组、银杏叶片组,对照组和Aβ组加入对照组血清,其他各组分别加入对应的含药血清,72 h后,Aβ组、脑还丹低剂量组、脑还丹高剂量组、银杏叶片组加入Aβ25-35作用72 h,测量各组海马细胞IL-1β、IL-6、TNF-α的mRNA水平,并检测海马神经元存活率。另外,用Aβ行立体定位制造痴呆大鼠模型,分组同上,分别给予生理盐水及高剂量脑还丹、低剂量脑还丹、银杏叶片治疗,1个月后取海马标本,观察海马区神经元超微结构。结果:(1)Aβ组海马细胞IL-1β、IL-6、TNF-α的mRNA水平明显高于对照组(P〈0.01),脑还丹高剂量组、脑还丹低剂量组和银杏叶片组海马细胞IL-1β、IL-6、TNF-α的mRNA水平均低于Aβ组(P〈0.01),其中以脑还丹高剂量组IL-1β、IL-6、TNF-α的mRNA水平最低。(2)Aβ组海马神经元存活率低于对照组(P〈0.01);脑还丹高剂量组、脑还丹低剂量组和银杏叶片组海马神经元存活率高于Aβ组(P〈0.01),其中以脑还丹高剂量组海马神经元存活率最高。(3)Aβ组大鼠海马区出现脂褐素堆积,溶酶体增大、空化,粗面内质网结构破坏,核糖体减少,线粒体减少,细胞核变形,突触减少和结构破坏。脑还丹组及银杏叶片组上述变化较Aβ组减轻。结论:脑还丹能通过减少炎性反应降低海马神经元损伤,从而改善神经退行性病变。Objective :To investigate the effect of Naohuan Dan(NHD) on inflammatory factors expression and ultrastructure of hippoeampus in dementia model rats induced by Aβ25-35. Methods:Cultured primary hippoeampal neurons were randomly divided into control group,15-amyloid protein(Aβ) group,NHD low-dose group,NHD high-dose group and ginkgo leaf tablet group. The control group and the Aβ group were added blank serum,and the other groups were added corresponding drug serum. After 72 hours,the Aβ group,NHD low-dose group,NHD high-dose group and ginkgo leaf tablet group were added Aβ25-35 for 72 hours. The expression of IL-1β mRNA,IL-6 mRNA,TNF-α mRNA and survival rates of hippoeampal neurons were mea-sured. In addition,the Aβ was used to induce dementia model rats and the model rats were divided into groups which were the same as before. The model rats in the control group and the Aβ group were given saline,in the NHD low-dose group were given low dose of NHD,in the NHD high-dose group were given high dose of NHD and in the ginkgo leaf tablet group were given ginkgo leaf tablet. The hippocampal sam-pies separated from the rats were observed after 1 month of treatment. Results:①Compared with the control group,the levels of IL-1β mRNA,IL-6 mRNA,TNF-et mRNA in the Aβ group were increased(P〈0.01),the levels of IL-1β mRNA,IL-6 mRNA,TNF-α mRNA in the NHD low-dose group,NHD high-dose group and ginkgo leaf tablet group were lower than those of the Aβ group(P〈0.01) and the levels of IL-1β mRNA,IL-6 mRNA,TNF-α mRNA in the NHD high-dose group were lower than all the other groups. (~)The survival rate of hippocampal neurons in the AI5 group was lower than that in the control group (P〈 0.01) , the survival rate of hippoeampal neurons in the NHD low-dose group,NHD high-dose group and ginkgo leaf tablet group was higher than that in the Aβ group(P〈0.01) and the survival rate in the NHD high-dose group was higher than all the other groups. ②Lipofuscin accumulation,lysosomes inc
